Description:   The DiO/DPA system detects cytoplasmic membrane potential changes using the principle of fluorescence resonance energy transfer (FRET), where DiO is a “stationary” FRET donor while DPA acts as a potentiometric FRET acceptor. The DiO/DPA system has been reported to produce a fluorescence signal change of >56% in HEK-293 cells and >25% in neuronal cultures and brain slices per 100 mV membrane potential change.

Description:   The Pax gene family of nuclear transcription factors is comprised of nine members that function during embryogenesis to regulate the temporal and position-dependent differentiation of cells. In addition, the family is involved in a variety of signal transduction pathways in the adult organism. Mutations in the Pax family of proteins have been linked to disease and cancer in humans. Pax-7 is a protein specifically expressed in cultured satellite cell-derived myoblasts. In situ hybridization reveals that Pax-7 is also expressed in satellite cells residing in adult muscle. A chromosomal aberration in the gene encoding Pax-7 causes rhabdomyosarcoma 2 (RMS2) (also called alveolar rhabdomyosarcoma).

Description:   PLAG1 is a transcription factor whose activation results in upregulation of target genes, such as IGFII, leading to uncontrolled cell proliferation: when overexpressed in cultured cells, higher proliferation rate and transformation are observed. Other target genes such as CRLF1, CRABP2, CRIP2, PIGF are strongly induced in cells with PLAG1 induction. Proto oncogene whose ectopic expression can trigger the development of pleomorphic adenomas of the salivary gland and lipoblastomas. Overexpression is associated with upregulation of IGFII, is frequently observed in hepatoblastoma, common primary liver tumor in childhood. Cooperates with CBFB-MYH11, a fusion gene important for myeloid leukemia.

Description:   Fibroblast growth factor-1 (FGF-1), also designated acidic FGF, and fibroblast growth factor-2 (FGF-2), also designated basic FGF, are members of a family of growth factors that stimulate proliferation of cells of mesenchymal, epithelial and neuroectodermal origin. Additional members of the FGF family include the oncogenes FGF-3 (Int2) and FGF-4 (hst/Kaposi), FGF-5, FGF-6, FGF-7 (KGF), FGF-8 (AIGF), FGF-9 (GAF) and FGF-10 through FGF-23. Members of the FGF family share 30-55% amino acid sequence identity and similar gene structure, and are capable of transforming cultured cells when overexpressed in trans- fected cells. Cellular receptors for FGFs are members of a second multigene family, including four tyrosine kinases designated Flg (FGFR-1), Bek (FGFR-L), TKF and FGFR-3.

Description:   A potent, cell permeable inhibitor of p38 MAP kinase (IC50=44nM for recombinant purified human p38α). Inhibits IL-1 and TNF-α production in LPS-stimulated human peripheral blood monocytes (IC50=120nM and 350nM, respectively). Reported to protect against mild excitotoxic neuronal injury caused by NMDA and provides substantial protection against cell death induced by either oxygen glucose deprivation (OGD) or magnesium deprivation in cultured neurons. Also, reduces myocardial reperfusion injury via inhibition of endothelial adhesion molecule expression and blockade of polymorphonuclear (PMN) accumulation.

Description:   Talin, a multifunctional constituent of cell-substratum attachment sites, is a high molecular weight protein (225-270 kDa) found in variety of tissues and cell types. It is localized at a subset of adherens junctions, specialized cell-cell and cell-matrix associations that are characterized by the presence of filamentous actin at the cytoplasmic face of the junctional complex. In cultured cells, talin is absent from cell-cell junctions and found predominantly at adhesion plaques and in fibrillar streaks underlying cell surface fibronectin. Talin interacts with at least two other proteins that are localized at adhesion plaques, vinculin and integrin. Talin and vinculin have been shown to interact with each other and both have been proposed to be involved in generating the transmembrane connection, between the extracellular matrix and the cytoskeleton, that occurs at adhesion plaques. At physiological ionic strength, talin is an elongate, flexible, monomeric protein with the ability to self-associate into dimers at higher protein concentrations.

Description:   Cyclins, regulatory subunits, which associate with kinases, control many of the important steps in cell cycle progression. The Cdc2 protein kinase (p34Cdc2) exhibits protein kinase activity in vitro and exists in a complex with both cyclin B and a protein homologous to p13SUC1. Cdc2 kinase is the active subunit of the M phase promoting factor (MPF) and the M phase-specific Histone H1 kinase. The p34Cdc2/cyclin B complex is required for the G2 to M transition. An additional cell cycle-dependent protein kinase, termed p55cdc, exhibits a high degree of homology with the S. cerevisiae proteins Cdc20 and Cdc4. The p55cdc transcript is readily detectable in a variety of cultured cell lines in growth phase, but disappears when cell growth is chemically arrested.

Description:   Adapalene, a synthetic naphthoic acid derivative, is a selective agonist of retinoic acid receptor (RAR)-β and RAR-γ (Kds = 34 and 130 nM, respectively). At 0,1-10 µM, adapalene dose-dependently inhibits sebocyte growth, proliferation and differentiation in a primary rat preputial cell culture system. Adapalene demonstrates anti-inflammatory and immunomodulatory activity by inhibiting 5-lipoxygenase-catalyzed leukotriene production and expression of toll-like receptor 2 on human monocytes. It is commonly used for the study of the pathogenesis and treatment of acne vulgaris.
