The cadherins are a family of Ca2+-dependent adhesion molecules that function to mediate cell-cell binding critical to the maintenance of structure and morphogenesis. Cadherins each contain a large extracellular domain at the N-terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. Cadherin-20, also known as protocadherin gamma B4, is a member of the protocadherin gamma gene cluster. Expressed primariy in fibroblasts, cadherin-20 plays a critical role in neuronal cell-cell connections in the brain. Through its ability to mediate cell-cell adhesion, caderin-20 is thought to help the body respond to injury by regulating post-traumatic wound healing.

Description:
This antibody recognizes a protein of ~35 kDa, identified as CD74 (Workshop IV). CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. Anti-CD74 has been shown to be useful in differentiating atypical fibroxanthoma (-) from malignant fibrous histiocytoma ( ).

LPP (LIM containing lipoma preferred partner), is a scaffolding protein which contains three LIM domains at its carboxy terminus, preceded by a proline rich pre LIM region containing a number of protein interaction domains. LPP localizes to sites of cell adhesion, such as focal adhesions and cell-cell contacts and may be involved in cell-cell adhesion and cell motility. LPP also shuttles through the nucleus and may function as a transcriptional co-activator. The human LPP gene maps to chromosomal location 3q28, and preferentially translocates to the HMGIC gene in a subclass of human benign mesenchymal tumors known as lipomas. Alternate splicing results in multiple transcript variants.

MHC class I antigens are heterodimers consisting of one alpha chain (44kDa) with beta 2 microglobulin (11.5 kDa). The antigen is expressed by all somatic cells at varying levels.
