Description:   In Western blotting, this antibody recognizes proteins in MW range of 265-400 kDa, identified as different glycoforms of EMA. This MAb reacts with the DTRP epitope in the tandem repeats. The α subunit has cell adhesive properties. It can act both as an adhesion and an anti-adhesion protein. EMA may provide a protective layer on epithelial cells against bacterial and enzyme attack. The β subunit contains a C-terminal domain, which is involved in cell signaling, through phosphorylations and protein-protein interactions. In immunohistochemical assays, it superbly stains routine formalin/paraffin carcinoma tissues. Antibody to EMA is useful as a pan-epithelial marker for detecting early metastatic loci of carcinoma in bone marrow or liver.

Description:   Lubricin, also designated proteoglycan-4 or megakaryocyte stimulating factor, is important for boundary lubrication within articulating joints. It is a disulfide-linked homodimer (between Cysteine 1146 and Cysteine 1403) that is essential for protein cleavage. Lubricin inhibits synovial cell adhesion to the cartilage surface, but also prevents the deposition of proteins from synovial fluid onto cartilage. Lubricin is highly expressed in cartilage, liver and synovial tissue. Defects in the gene encoding for lubricin can cause Jakobs syndrome, also designated camptodactyly-arthropathy-coxa vara-pericarditis syndrome (CACP). CACP is an autosomal recessive disorder characterised by joint failure associated with noninflammatory synoviocyte hyperplasia and subinitimal fibrosis of the synovial capsule. Lubricin undergoes different levels of glycosylation and may be detected at varying molecular weights.

Description:   Recognizes a protein of 42-46 kDa, identified as MAGE-1. This MAb does not cross-react with other members of MAGE-family. Human malignant neoplasms carry rejection antigens that are recognized by the patients' autologous, tumor directed and specific, cytolytic, CD8 T lymphocyte clones (CTL). The MAGE family of genes codes an important group of antigens. It was identified that melanomas and primary glial brain tumors express common melanoma associated antigens (MAAs). Because MAGE-1 is expressed on a significant proportion of human neoplasms of various histological types (melanoma, brain tumors of glial origin, neuroblastoma, non-small cell lung cancer, breast, gastric, colorectal, ovarian, renal cell carcinomas) and not on normal tissues, the encoded antigen may serve as a marker of early detection and target for cancer immunotherapy.

Description:   This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

Description:   IGF2BP2 (insulin-like growth factor 2 mRNA binding protein 2) is also known as IGF2 mRNA-binding protein 2, IMP-2 (IGF-II mRNA-binding protein 2), VICKZ family member 2 or hepatocellular carcinoma autoantigen p62 and is a 556 amino acid protein. IGF2BP2 is expressed in a variety of tissues including heart, placenta, skeletal muscle, pancreas, fetal liver, lung, kidney, thymus and gonadal cells. IGF2BP2 is an RNA binding protein which may be involved in the regulation of mRNA translation and may also function to control the spatial localization of target mRNAs. against IGF2BP2 have been detected in patients with HCC (hepatocellular carcinoma), suggesting that IGF2BP2 may have a role in the pathogenesis of HCC. Defects in IGF2BP2 are thought to be associated with susceptibility to type 2 diabetes mellitus.

Description:   CD17 is an intermediate glycosphingolipid from the metabolism of higher gangliosides that localizes to sphingolipid-sterol rafts. CD17 is detectable in monocytes, granulocytes, basophils, platelets, a subset of peripheral B cells (CD19 ) and tonsil dendritic cells. It is rapidly down regulated on activated granulocytes and is upregulated on IL-2 activated T lymphocytes. CD17 binds to bacteria and may function in phagocytosis. VEGF-treated endothelial cells can produce CD17, which can then mediate signaling toward PECAM-1 expression and angiogenesis. Tumor necrosis factor alpha (TNFa)-induced astrogliosis (astrocyte proliferation and glial fibrillary acidic protein (GFAP) upregulation) in response to neuro-inflammation (i.e. spinal cord injury) causes an increase in intracellular levels of CD17. Aberrant levels of glycosphingolipids are a feature of cancer cells and may influence integrin clustering and internalization.

Description:   Breast cancer metastasis-suppressor 1 (BRMS1) is 246 amino acid protein that acts as a mediator of metastasis suppression in several types of cancer including ovarian, lung, bladder, and murine mammary. BRMS1 mRNA is expressed in various tissues, including ovary, prostate, testis, and colon, but the protein is primarily detected in term placenta. BRMS1 suppresses metastasis without inhibiting tumorigenicity by modifying several metastasis-associated phenotypes. BRMS1 may participate in transcriptional regulation by binding to the mSin3/histone deacetylase complex. The expression of BRMS1 in certain cells increases connexin Cx43 expression and reduces connexin Cx32 expression. This produces a gap junction that increases intercellular communication, similar to those found in normal breast tissue. BRMS1 is stabilized by Hsp90 and may inhibit NF-â…¹B activity.

Description:   CDK5 (Cyclin Dependent Kinase 5) is serine/threonine kinase involved in synaptic regulation and neuronal development; phosphorylates synaptic protein Pctaire1; regulates acetylcholine receptor expression. CDK5 is a member of the cyclindependent kinase family of serine/threonine kinases. It is present in numerous mammalian tissues including kidney, testes, and ovary. Its activity is detected almost exclusively in brain extracts. Neuronal and muscle cells contain the highest amount of this protein. Similar to other Cdks, monomeric Cdk5 displays no enzymatic activity, but Cdk5 is not activated by cyclins. Instead, the p35 protein, which is expressed solely in the brain, activates Cdk5. Cdk5 interacts with D1 and D3 type G1 cyclins and can phosphorylate histone H1, TAU, MAP2 and NF-H and NF-M. Cdk5 activity is involved in terminal differentiation of neurons and muscle cells.

Description:   MuRF1, is a nuclear protein that interacts with SMT3b and the large myofibrillar protein Titin. In muscle cells, MuRF2 (RFN29) regulates gene expression and protein turnover. It localizes to the cytoplasm, but under atrophic conditions it is detected in the nucleus. MuRF2 can form oligomers with various other proteins, including Titin and Myosin. MuRF3, also designated tripartite motif-containing 54 (TRIM54) or ring finger protein 30 (RNF30), interacts with tubulin and stabilizes microtubules duing myotube formation. It is a cytoplasmic protein the localizes to the Z-lines in skeletal muscles, while MuRF2 localizes to the sarcomeric M-band in cardiomyocytes. MuRF3 shares 77% and 65% sequence identity with MuRF1 and MuRF2, respectively. MuRF1-3 share a conserved N-terminal RING domain and zinc-binding B-box motif, and two coiled-coil dimerization motif boxes, in their central regions.
