tumour necrosis factor receptor superfamily member 1A (Tnfrsf1a) is a member of the tumour necrosis factor receptor superfamily. Tnfrsf1a is one of the major receptors for the tumour necrosis factor-alpha. It can activate the transcription factor NF-κB, mediate apoptosis, and function as a regulator of inflammation. Antiapoptotic protein BCL2-associated athanogene 4 (BAG4/SODD) and adaptor proteins TRADD and TRAF2 have been shown to interact with this receptor, and thus play regulatory roles in the signal transduction mediated by the receptor. Germline mutations of the extracellular domains of this receptor were found to be associated with the human genetic disorder called tumour necrosis factor associated periodic syndrome (TRAPS) or periodic fever syndrome

UBE2M is a member of the E2 ubiquitin-conjugating enzyme family. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. This protein is linked with a ubiquitin-like protein, NEDD8, which can be conjugated to cellular proteins, such as Cdc53/culin. UBE2M accepts the ubiquitin-like protein NEDD8 from the UBA3-NAE1 E1 complex and catalyses its covalent attachment to other proteins. The specific interaction with the E3 ubiquitin ligase RBX1, but not RBX2, suggests that the RBX1-UBE2M complex neddylates specific target proteins, such as CUL1, CUL2, CUL3 and CUL4. It involved in cell proliferation and is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation.

Calcitonin is a secreted protein which belongs to the calcitonin family. Calcitonin is cleaved into the following two chains: Calcitonin and Katacalcin. Katacalcin is a potent plasma calcium-lowering peptide. Calcitonin is a 32-amino acid linear polypeptide hormone. Calcitonin acts to reduce blood calcium (Ca2+), opposing the effects of parathyroid hormone (PTH). Its importance in humans has not been as well established as its importance in other animals, as its function is usually not significant in the regulation of normal calcium homeostasis. Calcitonin causes a rapid but short-lived drop in the level of calcium and phosphate in blood by promoting the incorporation of those ions in the bones.

Mitochondrial Fission 1 Protein (FIS1) is a member of the FIS1 family. FIS1 is a single-pass membrane protein and contains one TPR repeat. FIS1 is part of the mitochondrial complex that promotes mitochondrial fission. FIS1 can induce cytochrome C discharge from the mitochondrion to the cytosol, eventually leading to apoptosis. In addition, FIS1 participates in peroxisomal growth and division. The C-terminus of FIS1 is required for mitochondrial or peroxisomal localization, while the N-terminus is necessary for mitochondrial or peroxisomal fission, localization and regulation of the interaction with DNM1L.

GHR is receptor for pituitary gland growth hormone involved in regulating postnatal body growth. On ligand binding, couples to the JAK2/STAT5 pathway. Adiponectin is a secreted protein. It is synthesised exclusively by adipocytes and secreted into plasma. Adiponectin is an important adipokine that is involved in the control of fat metabolism and insulin sensitivity, with direct anti-diabetic, anti-atherogenic and anti-inflammatory activities. Adiponectin Stimulates AMPK phosphorylation and activates in the liver and the skeletal muscle, enhancing glucose utilisation and fatty-acid combustion. Adiponectin also antagonises TNF-alpha by negatively regulating its expression in various tissues such as liver and macrophages, and also by counteracting its effects. It inhibits endothelial NF-kappa-B signaling through a cAMP-dependent pathway. Adiponectin may play a role in cell growth, angiogenesis and tissue remodeling by binding and sequestering various growth factors with distinct binding affinities, depending on the type of complex: LMW, MMW or HMW.

Tau proteins are proteins which contain four Tau/MAP repeats. They promote microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity. They are abundant in neurons of the central nervous system and are less common elsewhere, but are also expressed at very low levels in CNS astrocytes and oligodendrocytes. The tau proteins are the product of alternative splicing from a single gene that in humans is designated MAPT. When tau proteins are defective, and no longer stabilize microtubules properly, they can result in several neurodegenerative disorders such as Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy.

Macrophage migration inhibitory factor(MIF) is a secreted protein and belongs to the MIF family. MIF is an important regulator of innate immunity. The circulating MIF binds to CD74 on other immune cells to trigger an acute immune response. Hence MIF is classified as an inflammatory cytokine. Furthermore glucocorticoids also stimulate white blood cells to release MIF and hence MIF partially counter acts the inhibitory effects that glucocorticoids have on the immune system. Finally trauma activates the anterior pituitary gland to release MIF.

Description:
Mesothelin is a cell surface glycoprotein whose expression is limited to mesothelial cells of the serosa (pleura, pericardium, and peritoneum) and epithelial cells of the trachea, tonsils, fallopian tube, and kidneys. Mesothelin plays an important role in cell survival, proliferation, migration, invasion, tumour progression, and resistance to chemotherapy. The overexpression of mesothelin can activate NF-κB and signal transducer and activator of transcription 3 (Stat3), inhibit apoptotic signalling and TNF- alpha-induced apoptosis, and accelerate the G1–S transition. Mesothelin is also found overexpressed in various cancers, including malignant mesothelioma, pancreatic or ovarian carcinoma, sarcomas and in some gastrointestinal or pulmonary carcinomas. As a result of its limited expression in normal tissues, mesothelin has been reported as an ideal tumour-associated marker for the development of targeted therapy.

BAFF is mainly produced by innate immune cells such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells. T cells, activated B cells, some malignant B cells and also non-lymphoid cells like astrocytes, synoviocytes and epithelial cells can also produce BAFF. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation. Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of BAFF leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases such as Sjoegren syndrome, Rheumatoid arthritis (RA), Multiple sclerosis (MS) and Systemic Lupus Erythematosus (SLE). BAFF has also increased levels in some lymphoid cancers.

The protein specifically promotes pancreatic beta cell proliferation and beta cell mass expansion, thereby improving glucose tolerance. It promotes pancreatic beta cell proliferation without insulin resistance. Also it acts as a blood lipid regulator by regulating serum triglyceride levels and possibly by promoting ANGPTL3 cleavage. It interacts with ANGPTL3. It predominantly expressed in liver and also expressed in adipose tissues. The ability of the protein to induce pancreatic beta cell proliferation is promising in diabetes therapy. Betatrophin treatment could supply or replace insulin injections by increasing the number of insulin-producing cells in diabetes.
