The charged multivesicular body proteins, commonly designated CHMPs, belong to the vacuolar sorting protein family and function as chromatin-modifying proteins. CHMP1-6 are all components of ESCRT (endosomal sorting complex required for transport) I, II or III complexes. These complexes are crucial for sorting endosomal articles into multivesicular bodies (MVBs), and are also required for the formation of these bodies. CHMP2B, also known as CHMP2.5 or vacuolar protein-sorting-associated protein 2-2, is a 213 amino acid cytosolic protein. Widely expressed in brain, heart, skeletal muscle, small intestine, pancreas, lung, placenta and leukocytes, CHMP2B associates directly with CHMP2A and vps4 for the disassembly of the ESCRT-III complex. Defects in the gene encoding CHMP2B have been shown to cause chromosome 3-linked frontotemporal dementia (FTD3).

IRGM belongs to a family of interferon-gamma inducible GTPases, belonging to the GTPase superfamily, and are selectively induced by IFN-gamma or bacterial lipopolysaccharide (LPS) stimulation. IRGM is primarily expressed in all cells derived from B-cell lineages, and is highly expressed in macrophages following IFN-gamma stimulation. IRGM has been shown to be required for host defences against a broad range of intracellular pathogens. Specifically, IRGM deficient mice show defects with intracellular microbial killing, phagosome maturation and autophagy. More recently IRGM has been shown to inhibit baseline hematopoietic proliferation and is required for a normal hematopoietic stem cell response to chemical and infectious stimuli.

p107 is a tumor suppressor protein that appears to be involved in cell cycle regulation, as it is phosphorylated in the S to M phase transition and is dephosphorylated in the G1 phase of the cell cycle. Both the RB1 protein and the product of this gene can form a complex with adenovirus E1A protein and SV40 large T-antigen, with the SV40 large T-antigen binding only to the unphosphorylated form of each protein. In addition, both proteins can inhibit the transcription of cell cycle genes containing E2F binding sites in their promoters. Due to the sequence and biochemical similarities with the RB1 protein, it is thought that the protein encoded by this gene may also be a tumor suppressor. Two transcript variants encoding different isoforms have been found for this gene.

CCDC34 (coiled-coil domain containing 34), also known as Renal carcinoma antigen NY-REN-41, is a 373 amino acid protein that is expressed in testis, breast, lung, placenta, liver and small intestine. A translocation between the short arms of chromosomes 11 and 18 affecting the CCDC34 gene has been identified in a patient with hamartoma of the retinal pigment epithelium. There are two isoforms of CCDC34 that are produced as a result of alternative splicing events.

FBL3B is a 434 amino acid protein encoded by the human gene FBXL21. FBL3B contains one 40 amino acid F-box region, making it a member of the F-box family. FBL3B also contains three LRR (leucine-rich) repeats. RBMX2 is a 322 amino acid member of the IST3 family that contains one RRM (RNA recognition motif) domain. The RBMX2 gene is intronless, conserved in chimpanzee, dog, cow, mouse, rat, zebrafish, fruit fly, mosquito, C.elegans, S.pombe, S.cerevisiae, K.lactis, E.gossypii, M.grisea, N.crassa, A.thaliana, rice and P.falciparum, and maps to human chromosome Xq25. The X and Y chromosomes are the human sex chromosomes. Chromosome X consists of about 153 million base pairs and nearly 1,000 genes. The combination of an X and Y chromosome lead to normal male development while two copies of X lead to normal female development. More than one copy of the X chromosome with a Y chromosome causes Klinefelter's syndrome. A single copy of X alone leads to Turner's syndrome. More than 2 copies of the X chromosome, in the absence of a Y chromosome, is known as Triple X syndrome. Color blindness, hemophilia, and Duchenne muscular dystrophy are well known X chromosome-linked conditions that affect males more frequently as males carry a single X chromosome.
