Ephrin type-A receptor 4(EPHA4) belongs to the protein kinase superfamily and Ephrin receptor subfamily. EPHA4 contains 1 Eph LBD domain, 2 fibronectin type-III domains, 1 protein kinase domain and 1 SAM domain. EPH and EPH-related receptors have been implicated in mediating developmental events, particularly in the nervous system. Receptors in the EPH subfamily typically have a single kinase domain and an extracellular region containing a Cys-rich domain and 2 fibronectin type III repeats. CD99 Antigen-Like Protein 2 (CD99L2) belongs to the CD99 family. CD99L2 is a single-pass type I membrane protein and expressed in many tissues, with low expression in thymus. CD99L2 plays a role in a late step of leukocyte extravasation helping cells to overcome the endothelial basement membrane. CD99L2 and CD99 are involved in trans-endothelial migration of neutrophils in vitro and in the recruitment of neutrophils into inflamed peritoneum. A similar protein in mouse functions as an adhesion molecule during leukocyte extravasation. Alternate splicing results in multiple transcript variants.

Semaphorin-4D is also known as A8,BB18, GR3, CD100. Semaphorin-4D belongs to the semaphorin family containing 1 Ig-like C2-type domain, 1 PSI domain and 1 Sema domain. It is the cell surface receptor for PLXN1B and PLXNB2 that plays an important role in cell-cell signalling. It promotes the migration of cerebellar granule cells and of endothelial cells, regulates dendrite and axon branching and morphogenesis. Natural Cytotoxicity Triggering Receptor 3 (NCR3) along with NKp44 and NKp46 constitute a group of receptors termed “Natural Cytotoxicity Receptors”. They play a major role in triggering NK-mediated killing of most tumour cells lines. NKp30 is a type I transmembrane protein having a single extracellular V-like immunoglobulin domain. NKp30 is selectively expressed both in resting and activated human NK cells. In addition, NKp30 is also involved in NK-mediated induction of dendritic cell (DC) maturation. It has been demonstrated that NK cell activation signalling specifically induces lytic activity against several tumour cell types and synthesis of new NF-κB dependent proteins during the initiation of cytotoxicity.

T-Cell Surface Glycoprotein CD3 epsilon Chain (CD3 epsilon ) is a single-pass type I membrane protein. CD3 epsilon contains 1 Ig-like (immunoglobulin-like) domain and 1 ITAM domain. CD3 epsilon is a polypeptide encoded by the CD3E gene on chromosome 11 in humans. The T cell receptor-CD3 complex (TCR/CD3 complex) is involved in T-cell development and several intracellular signal-transduction pathways. This complex is critical for T-cell development and function, and represents one of the most complex transmembrane receptors. The T cell receptor-CD3 complex is unique in having ten cytoplasmic immunoreceptor tyrosine-based activation motifs (ITAMs). TCR/CD3 complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways.
