Making up nearly 6% of the human genome, chromosome 6 contains around 1200 genes within 170 million base pairs of sequence. Deletion of a portion of the q arm of chromosome 6 is associated with early onset intestinal cancer suggesting the presence of a cancer susceptibility locus. Porphyria cutanea tarda is associated with chromosome 6 through the HFE gene which, when mutated, predisposes an individual to developing this porphyria. Notably, the PARK2 gene, which is associated with Parkinson's disease, and the genes encoding the major histocompatiblity complex proteins, which are key molecular components of the immune system and determine predisposition to rheumatic diseases, are also located on chromosome 6. Stickler syndrome, 21-hydroxylase deficiency and maple syrup urine disease are also associated with genes on chromosome 6. A bipolar disorder susceptibility locus has been identified on the q arm of chromosome 6. The C6orf201 gene product has been provisionally designated C6orf201 pending further characterisation.

ACSF1 is a 672 amino acid protein belonging to the ATP-dependent AMP-binding enzyme family. Encoded by a gene that maps to human chromosome 12q24.31, ACSF1 is highly expressed in kidney, heart and brain, and shows similar neural expression as HMGCR (3-hydroxy-3-methylglutaryl-CoA reductase). Existing as three alternatively spliced isoforms, ACSF1 participates in ATP binding, ligase activity, acetoacetate-CoA ligase activity and nucleotide binding. The ACSF1 promoter is a known PPAR?target gene, with the nuclear receptor recruited to the ACSF1 promoter by direct interaction with stimulating protein-1 (Sp1). ACSF1 activates acetoacetate and is highly regulated by modulators that affect HMGCR and cholesterol biosynthesis.

HGFA Inhibitor 1 is an endogenous inhibitor of the kunitz-type serine proteinase HGF-activator (HGFA). HGF was first described as a hepatocyte-specific mitogen and survival factor, and has since been shown to exert a variety of actions on many cell types by binding to its MET receptor. HGF is activated by cleavage of the single-chain form to form a two-chain version by HGFA. HGFA Inhibitor 1 also inhibits trypsin, and MTSP-1 (Matriptase), a serine proteinase that also activates HGF. HGFB Inhibitor 1 is a splice variant of HGFA Inhibitor 1, and contains a 16 amino acid insert after the first kringle domain, relative to the HGFA Inhibitor 1 sequence.

During embryogenesis, HOX genes play a critical role in the spatial and temporal differentiation of cells. HoxC4, a sequence-specific transcription factor, belongs to the Antp HOX family and localizes to the nucleus. It functions as a part of a developmental regulatory system, providing cells with specific positional identities on the anterior-posterior axis. Expressed in brain, EF-CBP2 is suggested to bind metabotropic glutamate receptor 5 (mGluR-5) in a calcium-regulated manner. The gene encoding EF-CBP2 maps to human chromosome 16, which encodes over 900 genes and comprises nearly 3% of the human genome. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16, as is Crohn's disease, which is a gastrointestinal inflammatory condition.

BTBD14B (BTB/POZ domain-containing protein 14B), also known as NACC1 (nucleus accumbens associated 1), BEND8 or NAC1, is a 527 amino acid protein that localizes to both the nucleus and the cytoplasm and contains one BTB (POZ) domain. Existing as a homooligomer that interacts with HDAC3 and HDAC4, BTBD14B functions as a transcriptional repressor that influences the transcriptional activity of CRIF1 and is required for proteasome recruitment to the nucleus and cytoplasm in dendritic spines. BTBD14B is overexpressed in multiple carcinomas, suggesting a role in tumor development and metastasis.
