Src is the human homolog of the v-Src gene of the rous sarcoma virus, also designated avian sarcoma virus or ASV. Src was the first proto-oncogenic non-receptor tyrosine kinase characterized in human. The Src family, which has common structural motifs, is composed of nine members in vertebrates, including Src, Yes, Fgr, Frk, Fyn, Lyn, Hck, Lck and Blk. Src-family kinases transduce signals that are involved in the control of a variety of cellular processes, including proliferation, differentiation, motility and adhesion. Src-family ki-nases contain an amino-terminal cell membrane anchor followed by an SH3 domain and an SH2 domain, which are involved in modular association and activation, respectively. Src-family kinases, which are normally maintained in an inactive state and can be activated transiently during cellular events such as mitosis. Different subcellular localizations of Src-family kinases may be important for the regulation of specific cellular processes such as mitogenesis, cytoskeletal organization and membrane trafficking. c-Fgr is a human non-receptor tyrosine kinase family member that was discovered by using a probe toward the v-Fgr portion of the cell-derived domain of Gardner-Rasheed feline sarcoma virus. The human c-Fgr gene encodes a 529 amino acid protein.

The reticulon protein is a potent neurite outgrowth inhibitor which may also help block the regeneration of the central nervous system in higher vertebrates. Developmental neurite growth regulatory factor with a role as a negative regulator of axon-axon adhesion and growth, and as a facilitator of neurite branching. Reticulon 4 regulates neurite fasciculation, branching and extension in the developing nervous system. Involved in down-regulation of growth, stabilization of wiring and restriction of plasticity in the adult CNS. It regulates the radial migration of cortical neurons via an RTN4R-LINGO1 containing receptor complex. May inhibit BACE1 activity and amyloid precursor protein processing.

Syntaxins, a family of proteins involved in the fusion of synaptic vesicles with the plasma membrane, display broad tissue distribution and contain carboxy-terminal hydrophobic domains that direct themselves to their respective intracellular compartments. Synaptin 6 is a 255 amino acid protein that is widely expressed, with higher expression levels in brain, lung and kidney. This synaptin co-localizes with vesicle associated membrane protein (VAMP) 4 to tubular and vesicular membranes of the Golgi apparatus. The cytosolic domain of Syntaxin 6 reduces the rate on Glut4 reinternalization upon insulin withdrawl and is involved in a memrane-trafficking process that removes Glut4 from traffic directed to the plasma membrane. Syntaxin 6 is upregulated in activated macrophages in conjunction with an increase in the secretion of cytokines. The delivery of microdomain-associated lipids and proteins to the cell surface is regulated by Syntaxin 6.

C10orf28 is a 792 amino acid protein that exists as three alternatively spliced isoforms. The gene encoding C10orf28 maps to human chromosome 10, which spans nearly 135 million base pairs, makes up approximately 4.5% of total DNA in cells and encodes nearly 1,200 genes. Several protein-coding genes, including those that encode for chemokines, cadherins, excision repair proteins, early growth response factors (Egrs) and fibroblast growth receptors (FGFRs), are located on chromosome 10. Defects in some of the genes that map to chromosome 10 are associated with Charcot-Marie Tooth disease, Jackson-Weiss syndrome, Usher syndrome, nonsyndromatic deafness, Wolman’s syndrome, Cowden syndrome, multiple endocrine neoplasia type 2 and porphyria.
