Description:   The G protein-coupled receptor TGR5 is a 330-amino acid protein that is almost universally expressed in human tissues including heart, skeletal muscle, spleen, kidney, liver, small intestine, placenta, and leukocytes, but not in brain, colon (without mucosa), thymus, or lung. TGR5 is sensitive to bile acids and responds through a significant mechanism that coordinates energy homeostasis. Bile acids activate mitogen-activated protein (MAP) kinase pathways, specifically induce TGR5 internalization, promote an increase of guanosine 5'-O-3-thio-triphosphate binding in membrane fractions, and cause rapid intracellular cAMP production. Bile acids also provoke TGR5 to suppress macrophage functions. Binds to and phosphorylates RAD51 at 'Thr-309', which promotes the release of RAD51 from BRCA2 and enhances the association of RAD51 with chromatin, thereby promoting DNA repair by homologous recombination. Phosphorylates multiple sites within the C-terminus of TP53, which promotes activation of TP53 by acetylation and promotes cell cycle arrest and suppression of cellular proliferation. Also promotes repair of DNA cross-links through phosphorylation of FANCE. Binds to and phosphorylates TLK1 at 'Ser-743', which prevents the TLK1-dependent phosphorylation of the chromatin assembly factor ASF1A. This may enhance chromatin assembly both in the presence or absence of DNA damage. May also play a role in replication fork maintenance through regulation of PCNA.

Description:   Cenexin1 is an isoform of ODF2, that unlike ODF2 is present in several somatic cell types. Cenexin1 acts as a general scaffold protein that is specifically localised to the distal/subdistal appendages of mother centrioles. Cenexin1 is required for proper localization of Plk1 to the centrosomes. This centrosomal localization of Plk1 is required for proper microtubule function. Cenexin1 recruits Plk1 via a C-terminal extension of cenexin1 that is not present in ODF2. Cenexin1 is required for proper mitotic progression; depletion of Cenexin1 ultimately leads to chromosome missegregation and apoptosis. The ODF2 (outer dense fiber 2) gene encodes both ODF2 and Cenexin1, which have very different functions. ODF2 is a major component of sperm tail outer dense fibers (ODFs). ODFs are filamentous structures located on the outside of the axoneme in the midpiece and principal piece of the mammalian sperm tail. Description:   Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf146 gene product has been provisionally designated C1orf146 pending further characterization.
