MPP3, also known as Discs large homolog 3 (DLG3), is a 585 amino acid protein belonging to the MAGUK family. Proteins in the MAGUK family, including CASK, ZO-1, ZO-2, p55 and MPP2, are known to regulate cell proliferation, intracellular junctions and signaling pathways by interacting with the cytoskeleton. MPP3 contains one guanylate kinase-like domain, one PDZ domain, two L27 domains and one SH3 domain. Via the PDZ domain, MMP3 interacts with the C-terminus of TSLC1. MMP3 has also been found to interact with SR-2A and SR-4.

AKT, also known as protein kinase B (PKB), is a 57 kDa serine/threonine protein kinase. There are three mammalian isoforms of Akt: AKT1 (PKB alpha), AKT2 (PKB beta) and AKT3 (PKB gamma) with AKT2 and AKT3 being approximately 82% identical with the AKT1 isoform. PI3 kinase recruits AKT to the membrane, where it is activated by PDK1 phosphorylation. The Isocitrate dehydrogenase cytoplasmic enzyme is a homodimer of 416 residues that belongs to the isocitrate and isopropylmalate dehydrogenases family. IDHC catalyzes the third step of the citric acid cycle, which involves the oxidative decarboxylation of isocitrate, forming ?ketoglutarate and CO2 in a two step reaction. The first step involves the oxidation of isocitrate to the intermediate oxalosuccinate, while the second step involves the production of ?ketoglutarate. During this process, either NADH or NADPH is produced along with CO2. Ca2+ can bind to IDHC as a complex with isocitrate, acting as a competitive inhibitor of Mg2+. The IDHC enzyme is inactivated by phosphorylation at Ser-113 and contains a clasp-like domain wherein both polypeptide chains in the dimer interlock. IDHC is expressed in a wide range of species and also in organisms that lack a complete citric acid cycle.
