The early region (E1) of the adenovirus genome, responsible for transforming activity, is localised within the left most 11% of the viral genome and consists of two transcriptional units E1A and E1B. E1A is sufficient for partial transformation and immortalisation of primary cells. E1A gene products are necessary for normal levels of transcription of the other early regions of the adenovirus genome during productive infection and are able to either activate or repress the transcription of specific cellular genes. E1A forms specific complexes with cellular proteins including p105 causing inhibition of the cell cycle inducing arresting function of p105.

COMMD or COMM is a new family of proteins with homology to MURR1, a multifunctional protein that inhibits NFkB. These proteins form multimeric complexes and were identified in a biochemical screen for MURR1-associated factors. The family is defined by the presence of a conserved and unique motif termed the COMM (copper metabolism gene MURR1) domain, which functions as an interface for protein-protein interactions. The proteins designated as COMMD or COMM domain containing 1-10 are extensively conserved in multicellular eukaryotic organisms and define a novel family of structural and functional homologs of MURR1. The prototype of this family, MURR1/COMMD1, suppresses NFkB not by affecting nuclear translocation or binding of NF-kappaB to cognate motifs; rather, it functions in the nucleus by affecting the association of NF-kappaB with chromatin.

PHF2 is a 1,101 amino acid protein belonging to the PHD finger protein family. Members of the PHD finger protein family function as transcriptional regulators that affect gene expression by modulating chromatin structure. Expressed in a wide variety of tissues, PHF2 localizes to the nucleus and contains one PHD-type zinc finger and one JMJC domain, suggesting a possible role for PHF2 in transcription regulation and chromatin remodeling. The gene encoding PHF2 lies in the candidate region for hereditary sensory neuropathy type I (HSN1), a disorder characterized by sensory dysfunction. PHF2 exhists as two isoforms produced by alternative splicing, designated isoform 1 and 2 respectively.

Adipogenesis, the process of transforming pre-adipocytes into mature fat cells, is of particular interest due to the role adipocytes play in obesity and type II diabetes. Adipocytes have been shown to affect a variety of functions, including hemostasis, angiogenesis and energy balance, by secreting hormones and bioactive peptides. The FNDC3B protein, also designated FAD104 (factor for adipocyte differentiation 104) or HCV NS5A-binding protein 37, is expressed during early adipogenesis. Belonging to the FNDC3 family of proteins, FNDC3B is a 1,204 amino acid protein that contains nine fibronectin type-III domains. FNDC3B-deficient mice die within one day of birth, suggesting that FNDC3B is crucial for postpartum survival. Mouse embryonic fibroblasts (MEFs) with loss of FNDC3B function displayed a reduction in stress fiber formation, indicating a role for FNDC3B in cell proliferation, adhesion, spreading and migration.
