Description:   The c Abl proto oncogene encodes a protein tyrosine kinase that is located in the cytoplasm and nucleus. In chronic myelogenous leukaemia and in a subset of acute lymphoblastic leukaemias, the c Abl proto oncogene undergoes a (9;22) chromosomal translocation producing a novel rearranged chromosome (the Philadelphia chromosome) As the result of the fusion of c Abl sequences from chromosome 9 to the Bcr gene on chromosome 22. The molecular consequence of this translocation is the generation of a chimeric Bcr/Abl mRNA encoding activated Abl protein tyrosine kinase.

Description:   Nuclear MarkerRb is a tumor suppressor gene which functions as a negative regulator of the cell cycle by interacting with transcription factors including E2F1, PU1, ATF2, UBF, Elf1 and cAbl. This ability of Rb to alter transcription is regulated by phosphorylation catalyzed by the cyclin dependent protein kinases (cdks). Rb is phosphorylated on serine and threonine, but not on tyrosine residues. It forms a complex with SV40 large T antigen, adenovirus E1A, and human papilloma virus 16E. Rb protein may act by regulating transcription and loss of its function leads to uncontrolled cell growth. Aberrations in the Rb gene have been implicated in cancers of breast, colon, prostate, kidney, nasopharynx, and leukemia.

Description:   Epstein-Barr virus-induced gene 3 (Ebi3) is a widely expressed homolog to the interleukin IL-12 p40 subunit protein that forms a heterodimer with either IL-12 p35 or an IL-12 p35 homolog, p28, to create a new cytokine (IL-27). Ebi3 may function to antagonize IL-12 and to inhibit the development of a Th1 immune response. Ebi3 is strongly expressed in Hodgkin and Reed-Sternberg cells, independently of the EBV status of the tumor cells. Ebi2 is expressed in B-lymphocytes and lymphoid tissues and may function in the modulation of the immune system. Out of the nine genes that are induced by the Epstein-Barr virus, Ebi2 exhibits the highest levels of up-regulation. In addition, Ebi2 may play a role mediating normal lymphocyte functions.

Description:   Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. The protein encoded by this gene is one of the PKC family members. This kinase has been shown to be involved in many different cellular functions, such as neuron channel activation, apoptosis, cardioprotection from ischemia, heat shock response, as well as insulin exocytosis. Knockout studies in mice suggest that this kinase is important for lipopolysaccharide (LPS)-mediated signaling in activated macrophages and may also play a role in controlling anxiety-like behavior. Considered to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterised by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. Chromosome 9 is partnered with chromosome 22 in the translocation leading to the aberrant production of BCR-ABL fusion protein often found in leukemias. The C9orf68 gene product has been provisionally designated C9orf68 pending further characterisation. There are two isoforms of C9orf68 that are produced as a result of alternative splicing events.

Description:   Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf113 gene product has been provisionally designated C1orf113 pending further characterization.

Description:   The protein encoded by this gene is part of a complex of proteins that constitute adherens junctions (AJs). AJs are necessary for the creation and maintenance of epithelial cell layers by regulating cell growth and adhesion between cells. The encoded protein also anchors the actin cytoskeleton and may be responsible for transmitting the contact inhibition signal that causes cells to stop dividing once the epithelial sheet is complete. Finally, this protein binds to the product of the APC gene, which is mutated in adenomatous polyposis of the colon. Mutations in this gene are a cause of colorectal cancer (CRC), pilomatrixoma (PTR), medulloblastoma (MDB), and ovarian cancer. Description:   In all vertebrates, SV2 proteins are abundant, hydrophobic, membrane glycoproteins that are expressed as two major isoforms, SV2A and SV2B, and one minor isoform, SV2C. SV2 proteins are differentially expressed in the brain and are present on all synaptic vesicles, independent of transmitter type. SV2A is abundantly expressed in the subcortex, specifically in the synaptic vesicles of all presynaptic nerve terminals, and also in most neuroendocrine secretory granules. SV2B displays a more restricted pattern of expression in that it is only present on a small subset of synapses in the hippocampus and cortex. SV2A and SV2B are functionally redundant and are required for maintaining normal brain function in vertebrates. SV2A and SV2B mediate synaptic transmission by regulating cytoplasmic Ca²⁺ levels in the nerve terminal during repetitive stimulation.
