Calmodulin is a primary mediator of Ca2+ signals in the nervous system. Ric, a protein related to the Ras subfamily of small GTPases, has the ability to bind calmodulin. In addition, two Ras-like human proteins, Rin and Rit (Ric-related gene expressed in many tissues), which are 71% and 66% identical to RIC respectively, share related G2 domains with Ric. While most members of the Ras subfamily are plasma membrane-associated and generally require a C-terminal isoprenyl group to bind to the plasma membrane, Rit and Rin lack the recognition signal for C-terminal prenylation. Transiently expressed Rit and Rin are plasma membrane-localized because both proteins contain a C-terminal cluster of basic amino acids, which provides a mechanism for membrane association. Rin binds calmodulin through a C-terminal binding motif. Rit and Ric are widely expressed, whereas expression of Rin is restricted to the neuron system. Description:   Sulfation is an essential conjugation reaction that increases the water solubility of many compounds, thereby influencing their renal excretion and also resulting in the formation of active metabolites. SULT4A1 (sulfotransferase family 4A, member 1), whose alternative names include brain sulfotransferase-like protein, nervous system sulfotransferase, NST, SULTX3, hBR-STL-1, BRSTL1, BR-STL-1, MGC40032 and DJ388M5.3, is a 284 amino acid protein showing cytoplasmic localization. As a member of the sulfotransferase 1 family, SULT4A1 plays a role in elimination of xenobiotics, activation of procarcinogens and regulation of hormones. SULT4A1 is highly expressed in cerebral cortex and frontal lobe, with lower expression in cerebellum, temporal and occipital lobes. Two SULT4A1 isoforms exist to alternative splicing events. The gene encoding SULT4A1 maps to human chromosome 22q13.2, a region which has been implicated in predisposition to schizophrenia.

Description:   Retinoids are metabolites of vitamin A (retinal) and are believed to represent important signaling molecules during vertebrate development and tissue differentiation. Two families of retinoid receptors have been identified. Retinoic acid receptors (RARs) include RAR alpha, RAR beta and RAR gamma, each of which has a high affinity for all trans retinoic acids and belongs to the same class of nuclear transcription factors as thyroid hormone receptors, vitamin D3 receptor and ecdysone receptor. The ligand binding domains of the RARs are highly conserved and RAR isoforms are expressed in distinct patterns through out development and in the mature organism. Description:   Members of the calsyntenin protein family are localized to the post-synaptic membrane of exicitatory central nervous system (CNS) synapses. Calsyntenin-2, also known as Alcadein-gamma, is a 955 amino acid protein that localizes to the endoplasmic reticulum, golgi apparatus and plasma membranes. Containing 2 cadherin-like repeats in its N-terminal extracellular region, calsyntenin-2 binds synaptic calcium with its cytoplasmic domain, suggesting a role in the modulation of calcium-mediated postsynaptic signals. Under normal physiological conditions, calsyntenin-2 is protoeolytically processed in an event in which the primary zeta-cleavage generates a short C-terminal transmembrane fragment and a long extracellular N-terminal domain.

Description:   Chromosome 7 is about 158 milllion bases long, encodes over 1000 genes and makes up about 5% of the human genome. Chromosome 7 has been linked to Osteogenesis imperfecta, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome. The deletion of a portion of the q arm of chromosome 7 is associated with Williams-Beuren syndrome, a condition characterized by mild mental retardation, an unusual comfort and friendliness with strangers and an elfin appearance. Deletions of portions of the q arm of chromosome 7 are also seen in a number of myeloid disorders including cases of acute myelogenous leukemia and myelodysplasia.

Description:   Nuclear lamins form a network of intermediate-type filaments at the nucleoplasmic site of the nuclear membrane. Two main subtypes of nuclear lamins can be distinguished, i.e. A type lamins and B type lamins. The A type lamins comprise a set of three proteins arising from the same gene by alternative splicing, i.e. lamin A, lamin C and lamin Adel 10, while the B type lamins include two proteins arising from two distinct genes, i.e. lamin B1 and lamin B2. Recent evidence has revealed that mutations in A-type lamins give rise to a range of rare but dominant genetic disorders, including Emery-Dreifuss muscular dystrophy, dilated cardiomyopathy with conduction-system disease and Dunnigan-type familial partial lipodystrophy. In addition, the expression of A type lamins coincides with cell differentiation and as A type lamins specifically interact with chromatin, a role in the regulation of differential gene expression has been suggested for A type lamins.
