Description:   Functions as a transcriptional activator in a promoter context-dependent manner. Modulates the placental expression of CYP11A1. Involved in regulation of the alpha-globin gene in erythroid cells. Activation of the alpha-globin promoter in erythroid cells is via synergistic interaction with TFCP2 (By similarity). Involved in regulation of the alpha-globin gene in erythroid cells. Binds strongly to sequences around the HIV-1 initiation site and weakly over the TATA-box. Represses HIV-1 transcription by inhibiting the binding of TFIID to the TATA-box.

Description:   This gene is a member of the UBA domain family, whose members include proteins having connections to ubiquitin and the ubiquitination pathway. The ubiquitin associated domain is thought to be a non-covalent ubiquitin binding domain consisting of a compact three helix bundle. This particular protein originates from a gene locus in a refined region on chromosome 9 undergoing loss of heterozygosity in nasopharyngeal carcinoma (NPC). Taking into account its cytogenetic location, this UBA domain family member is being studied as a putative target for mutation in nasopharyngeal carcinomas. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene.

Description:   NF-kappa-B is a pleiotropic transcription factor present in almost all cell types and is the endpoint of a series of signal transduction events that are initiated by a vast array of stimuli related to many biological processes such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis. NF-kappa-B is a homo- or heterodimeric complex formed by the Rel-like domain-containing proteins RELA/p65, RELB, NFKB1/p105, NFKB1/p50, REL and NFKB2/p52 and the heterodimeric p65-p50 complex appears to be most abundant one. The dimers bind at kappa-B sites in the DNA of their target genes and the individual dimers have distinct preferences for different kappa-B sites that they can bind with distinguishable affinity and specificity. Different dimer combinations act as transcriptional activators or repressors, respectively. NF-kappa-B is controlled by various mechanisms of post-translational modification and subcellular compartmentalization as well as by interactions with other cofactors or corepressors. NF-kappa-B complexes are held in the cytoplasm in an inactive state complexed with members of the NF-kappa-B inhibitor (I-kappa-B) family. In a conventional activation pathway, I-kappa-B is phosphorylated by I-kappa-B kinases (IKKs) in response to different activators, subsequently degraded thus liberating the active NF-kappa-B complex which translocates to the nucleus. NF-kappa-B heterodimeric p65-p50 and p65-c-Rel complexes are transcriptional activators. The NF-kappa-B p65-p65 complex appears to be involved in invasin-mediated activation of IL-8 expression. The inhibitory effect of I-kappa-B upon NF-kappa-B the cytoplasm is exerted primarily through the interaction with p65. p65 shows a weak DNA-binding site which could contribute directly to DNA binding in the NF-kappa-B complex. Associates with chromatin at the NF-kappa-B promoter region via association with DDX1. Essential for cytokine gene expression in T-cells (PubMed:15790681).

Description:   NGL-1 is a single pass type I membrane protein that acts as a cell adhesion molecule. It contains nine leucine-rich repeats (LRR) and one Ig-like C2-type domain. NGL-1 is predominantly expressed in the striatum and the cerebral cortex of both the embryonic and adult brain. NGL-1 specifically interacts with Netrin G1 (a molecule involved in axon guidance in the developing central nervous system) via its LRR region. NGL-1 plays a role in the regulation of neurite outgrowth of developing thalamic neurons. Soluble NGL-1 inhibits thalamic axon outgrowth while NGL-1 that is bound to the surface of developing thalamocortical axons stimulates growth. NGL-1 also interacts with Whirlin possibly stablizing interstereociliar links.

Description:   NF-kappa-B is a ubiquitous transcription factor involved in several biological processes. It is held in the cytoplasm in an inactive state by specific inhibitors. Upon degradation of the inhibitor, NF-kappa-B moves to the nucleus and activates transcription of specific genes. NF-kappa-B is composed of NFKB1 or NFKB2 bound to either REL, RELA, or RELB. The most abundant form of NF-kappa-B is NFKB1 complexed with the product of this gene, RELA. Four transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2011].

Description:   PFKFB2 is involved in both the synthesis and degradation of fructose-2,6-bisphosphate, a regulatory molecule that controls glycolysis in eukaryotes. It has a 6-phosphofructo-2-kinase activity that catalyzes the synthesis of fructose-2,6-bisphosphate, and a fructose-2,6-biphosphatase activity that catalyzes the degradation of fructose-2,6-bisphosphate. PFKFB2 regulates fructose-2,6-bisphosphate levels in the heart, while a related enzyme encoded by a different gene regulates fructose-2,6-bisphosphate levels in the liver and muscle. This enzyme functions as a homodimer. There are two named isoforms.

Description:   Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamate-gated, cation-specific ion channels. Synaptic and extrasynaptic NMDA receptors have been shown to have opposite effects on neuronal survival, CREB function and gene regulation. As one of the four major proteins of the NMDA receptor ion channel, GRINA (Glutamate [NMDA] receptor-associated protein 1), also designated NMDA receptor glutamate-binding subunit or putative MAPK-activating protein PM02, is a 371 amino acid multi-pass transmembrane protein. Due to the chromosomal location of the gene encoding GRINA, studies have linked possible GRINA involvement with a form of idiopathic generalized epilepsy.

Description:   The temporal genetic hierarchy influencing normal limb development can deregulate and mediate mammalian developmental syndromes. In mice, the limb deformity (ld) locus influences normal limb development and gives rise to alternative mRNAs that can translate into a family of proteins known as formins. Formins play a crucial role in cytoskeletal reorganization by influencing Actin filament assembly. Formins co-localize with the actin cytoskeleton and can translocate into the cell cytosol and into the nucleus in an HGF-dependent manner. Vertebrate nuclear formins can control polarizing activity in limb buds through establishment of a Sonic hedgehog/FGF-4 feedback loop. Deficiency mutations at the mammalian ld locus lead to profound developmental defects in limb and kidney formation. The human Formin 1 and 2 genes map to chromosome 15q13.3 and 1q43, respectively.
