Belonging to the F-box family of proteins, FBXO45 is a 286 amino acid protein that contains one C-terminal F-box domain. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein)-type E3 ubiquitin ligase complex and are involved in substrate recognition and protein recruitment for ubiquitination. They are members of a larger family of proteins that are involved in the regulation of a wide variety of cellular mechanisms, including the cell cycle, the immune response, signaling cascades and developmental processes. They function by targeting proteins, such as cyclins, cyclin-dependent kinase inhibitors, I˚B-å and ∫-catenin, for degradation by the proteasome after ubiquitination. Via its F-box domain, FBXO45 can directly interact with Skp1 p19 and CUL-1. FBXO45 has been shown to be an estrogen-induced gene.

Glucose-6-phosphatase (G6Pase), is a multicomponent enzyme system that hydrolyzes glucose-6-phosphate in the final step of gluconeogenesis and gluconeolysis. G6Pase localizes to the endoplasmic reticulum, and while liver, kidney, and intestine are the only tissues that express the first identified isoform, G6Pase-alpha, a second form, designated G6Pase-∫, contributes to blood glucose homeostasis in a wider range of tissues. Islet-specific G-6-Pase catalytic subunit-related protein (IGRP), a homolog of the catalytic subunit of G6Pase, may play a role in the regulation of islet metabolism and in insulin secretion induced by metabolites. The exact catalytic acivity of IGRP is not defined. Identification of inhibitors of IGRP have potential therapeutic benefits for treatment of type 2 diabetes resulting from insulin secretion defects. Structurally, IGRP has been shown to be a glycoprotein held in the endoplasmic reticulum by nine transmembrane domains, which are then degraded in cells through the proteasome pathway generating MHC class I presented peptides.

Multiple isoforms of MARCH9 exist due to alternative splicing events.

The Six proteins (sine oculis) are a family of homeodomain transcription factors that share a conserved DNA binding domain. Six2, Six4 (AREC3) and Six5 bind to the same DNA sequence, indicating that they may regulate the same target genes.
