Cytokeratins are proteins of keratin-containing intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. The cytokeratins are encoded by a family encompassing 30 genes. Among them, 20 are epithelial genes and the remaining 10 are specific for trichocytes.
In the cytoplasm, the keratin filaments conform a complex network which extends from the surface of the nucleus to the cell membrane. Numerous accessory proteins are involved in the genesis and maintenance of such structure. This association between the plasma membrane and the nuclear surface provides important implications for the organization of the cytoplasm and cellular communication mechanisms. Apart from the relatively static functions provided in terms of supporting the nucleus and providing tensile strength to the cell, the cytokeratin networks undergo rapid phosphate exchanges mediated depolymerization, with important implications in the more dynamic cellular processes such as mitosis and post-mitotic period, cell movement and differentiation. Cytokeratins interact with desmosomes and hemidesmosomes, thus collaborating to cell-cell adhesion and basal cell-underlying connective tissue connection.

Casein kinase I (also designated CKI) and casein kinase II (CKII) compose a family of serine/threonine protein kinases which are present in all eukaryotes examined to date. Casein kinase I family members, which include casein kinase I Alpha, I Gamma, I Delta and I Epsilon, have been implicated in the control of cytoplasmic and nuclear processes, including DNA replication and repair, membrane trafficking, circadian rhythm, cell cycle progression, chromosome segregation, apoptosis and cellular differentiation. Casein kinase I isoform alpha-like (CSNK1A1L) is a 337 amino acid protein that shares a high degree of sequence similarity with the alpha isoform of casein kinase 1. CSNK1A1L resides in the cytoplasm and participates in the Wnt signaling pathway. By utilizing ATP within its protein kinase domain, CSNK1A1L phosphorylates a large number of proteins.

The Ovo family of zinc-finger transcription factors encode evolutionarily conserved genes including those from Caenorhabditis elegans, Drosophila melanogaster, mouse and human. Members of the Ovo family include Ovol1 and Ovol2. Ovol1 acts as a transcriptional repressor by interacting with key developmental signaling pathways such as Wnt and TGF-∫/BMP. Specifically, Ovol1 represses c-Myc and Id2 genes and establishes a balance between proliferation and differentiation of progenitor cells. Deletion of Ovol1 in mice leads to germ cell degeneration and defective sperm production in adult males. Ovol1 has also been shown to repress itself as well as Ovol2, which is thought to regulate neural development and vascular angiogenesis during embryogenesis.

Phocein is a 225 amino acid protein encoded by the human gene MOBKL3. Phocein belongs to the MOB1/phocein family and is phosphorylated on serine residues. Phocein is a widely expressed, highly conserved intracellular protein. The sequence of Phocein has limited homology to the sigma subunits from Clathrin adaptor complexes and contains an additional stretch bearing a putative SH3-binding domain. Phocein is usually associated with membranes but can be present in the cytosol, where it behaves as a protein complex. Phocein is the major partner of the striatin family members, which are scaffolding proteins involved in signaling and trafficking. Due to its association with Dynamin via direct interactions with nucleotide diphosphate kinase (NDPK) and Eps15, Phocein has been implicated in vesicular trafficking, acting in particular in the endocytic process.

Mapping to chromosome 21, the FASP1 gene (FAPP1-associated protein 1) encodes a 233 amino acid protein that is homologous to the fission yeast protein Mis18. In yeast, Mis18 is localized to the centrosome and forms a complex with Mis16 to maintain the deacetylated state of histones specifically in the central core of centromeres. FASP1, also known as Protein Mis18-alpha and C21orf45, is required for the recruitment of CENP-A to centrosomes and is thereby essential for normal chromosome segregation during mitosis. With expression in testis, FASP1 exists as a homodimer, a heterodimer with MIS18B or is present in a complex containing other Mis18 family members. FASP1 has been shown to bind to pp5644 in Hela cells, where overexpression of pp5644 leads to inhibited growth and colony formation.
