The homeobox DNA-binding domain is a 60 amino acid motif that is conserved among many species and functions to bind DNA via a helix-turn-helix structure, thereby playing a role in transcriptional regulation and the control of gene expression. HMX2 (H6 family homeobox 2), also known as H6L or Nkx5-2, is a 273 amino acid nuclear protein that belongs to the HMX homeobox family and contains one homeobox DNA-binding domain. HMX2 functions as a transcription factor that assists in specification of neuronal cell types and is essential for proper development of hypothalamus and inner ear. Hemizygous deletions of the gene encoding HMX2 are thought to lead to vestibular dysfunction, inner ear malformations and congenital sensorineural hearing loss.

TDE1, tumor differentially expressed protein 1, is a multi-pass membrane protein also known as serine incorporator 3. Also found on the membranes of the Golgi apparatus within cells, TDE1 is highly expressed in neuronal populations but is also found in thymus, kidney, liver and testis. Expression levels of TDE1 in tumors can be as much as tenfold the amount found in normal tissue of the same type. This increased expression implicates TDE1 as being involved in the cellular transformation from normal to malignant tissue. It is believed TDE1 contributes to oncogenesis by partially protecting cells from serum starvation and etoposide-induced apoptosis. The mechanism through which TDE1 protects cells is poorly understood, but may involve aberrant methylation of TDE1 complexes.

Cysteine-rich secretory proteins (CRISPs) represent a family of evolutionarily conserved proteins which play a role in the innate immune system and are transcriptionally regulated by androgens in several tissues. CRISP-8 (Cysteine-rich secretory protein 8), also known as PI15 (Peptidase inhibitor 15), P25TI or SugarCrisp, is a 258 amino acid secreted protein that belongs to the CRISP family. Expressed at low levels in thyroid, prostate, salivary and mammary tissue, CRISP-8 functions as a serine protease inhibitor that exhibits weak inhibitory action against Trypsin, a serine protease found in the digestive system. In addition to its role as a protease inhibitor, CRISP-8 is secreted in neuroblastoma and glioblastoma cell lines, suggesting a role for CRISP-8 in tumour formation and metastasis within the central nervous system.
