CYP2S1 is a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum. In rodents, the homologous protein has been shown to metabolize certain carcinogens; however, the specific function of the human protein has not been determined.

Description:
The immunophilins are a highly conserved family of cis-trans peptidyl-prolyl isomerases that bind to and mediate the effects of immunosuppressive drugs, such as cyclosporin, FK506 and rapamycin (1). Several related immunophilins, FKBP12, FKBP51 and FKBP52, are characterized as cytosolic FK506-binding proteins, and following ligand binding, they functionally inhibit the phosphatase activity of calcineurin (2,3). The ubiquitously expressed FKBP12 also associates with the cytoplasmic domain of the TGF?type I receptor, where it stabilizes the inactive conformation of the receptor and blocks the activation of the TGF?pathway (4). FKBP51 and FKBP52 are two highly related proteins (5,6). FKBP51 is predominantly expressed in T cells and is induced by glucocorticoids (5). FKBP51 mediates the effects of FK506 and rapamycin by inhibiting intracellular calcineurin activity, and by blocking T-cell activation and proliferation (7). FKBP52, known also as FKBP-59 or heat shock protein 56, is expressed in a variety of tissues and can also associate with the heat shock protein (hsp90) in mature steroid receptor complexes (6,8).

Rab17 belongs to the Rab family of small Ras-like GTPases. It is specifically expressed in epithelial cells and is upregulated during cell polarization. Immunofluorescence staining studies indicate that Rab17 is associated with the perinuclear recycling endosome in nonpolarized epithelial cells and with the apical recycling endosome in polarized epithelial cells. The function of Rab17 remains unclear. Reports of Rab17 colocalization with internalized IgA in the apical endosome suggest that it may regulate receptor-mediated transcytosis. Rab17 has also been shown to regulate melanocytic filopodia formation and melanosome trafficking. siRNA knockdown of Rab17 in melanoma cells induces melanosome accumulation in the cell periphery.

In chronic myelogenous leukemia and in a subset of acute lymphoblastic leukemias, the c Abl proto oncogene undergoes a (9;22) chromosomal translocation producing a novel rearranged chromosome (the Philadelphia chromosome) As the result of the fusion of c Abl sequences from chromosome 9 to the Bcr gene on chromosome 22. The molecular consequence of this translocation is the generation of a chimeric Bcr/Abl mRNA encoding activated Abl protein tyrosine kinase.

As one of the four major proteins of the NMDA receptor ion channel, GRINA (Glutamate [NMDA] receptor-associated protein 1), also designated NMDA receptor glutamate-binding subunit or putative MAPK-activating protein PM02, is a 371 amino acid multi-pass transmembrane protein. Due to the chromosomal location of the gene encoding GRINA, studies have linked possible GRINA involvement with a form of idiopathic generalized epilepsy.
