The ABL1 protooncogene encodes a cytoplasmic and nuclear protein tyrosine kinase that has been implicated in processes of cell differentiation, cell division, cell adhesion, and stress response. Activity of c-Abl protein is negatively regulated by its SH3 domain, and deletion of the SH3 domain turns ABL1 into an oncogene. The t(9;22) translocation results in the head-to-tail fusion of the BCR (MIM:151410) and ABL1 genes present in many cases of chronic myelogeneous leukemia. The DNA-binding activity of the ubiquitously expressed ABL1 tyrosine kinase is regulated by CDC2-mediated phosphorylation, suggesting a cell cycle function for ABL1. iASPP (inhibitor of apoptosis stimulating protein of p53, RelA-associated inhibitor (RAI), NFKap B-interacting protein 1, protein phosphatase 1 regulatory subunit 13-like, PPP1R13L) is the third member of the ASPP family of proteins. Unlike ASPP1 and ASPP2, which interact with p53 and enhance its ability to induce apoptosis, iASPP inhibits apoptosis induced by p53 overexpression. iASPP has been identified in a yeast-two hybrid screen as an interacting protein of the p65 subunit of NFKap B (RelA),3 interacts with NFKap B in vivo, and acts as an efficient inhibitor of HIV-1 gene expression in which both NFKap b and Sp1 play major roles. iASPP expression is upregulated in human breast carcinomas expressing wild-type p53, and gene overexpression may play an important role in the leukemogenesis and/or progression of acute leukemia. A crucial regulator of chromatin dynamics and DNA transcription is the covalent modification and methylation of histones. Generally, methylation of certain lysine residues on Histone H3 and Histone H4 can be associated with transcriptionally active or inactive chromatin. This regulation has profound effects on the expression of genes and is part of an epigenetic memory network that determines cell fate. JMJD7 (Jumonji domain-containing protein 7) is a member of a family of JMJC domain-containing histone demethylases that are directly involved in removing methyl residues from distinct and unique lysine residues. These actions are implicated in gene expression and the regulation of cell senescence. JMJC domain-containing histone demethylases are also likely involved in development via their ability to regulate gene expression. JMJD7 contains one JMJC domain and was originally thought to be an alternatively spliced isoform of PLA2G4B.

This gene encodes a protein that shares a high degree of sequence similarity with p21-activated kinase (PAK) family members. The proteins of this family are Rac/Cdc42-associated Ste20-like Ser/Thr protein kinases, characterised by a highly conserved amino-terminal Cdc42/Rac interactive binding (CRIB) domain and a carboxyl-terminal kinase domain. PAK kinases are implicated in the regulation of a number of cellular processes, including cytoskeleton rearrangement, apoptosis and the MAP kinase signaling pathway. The protein encoded by this gene was found to interact with androgen receptor (AR), which is a steroid hormone-dependent transcription factor that is important for male sexual differentiation and development. The p21-activated protein kinase 6 gene was found to be highly expressed in testis and prostate tissues and the encoded protein was shown to cotranslocate into the nucleus with AR in response to androgen.

PIRT is a 137 amino acid multi-pass membrane protein. Highly conserved among vertebrates, PIRT consists of two transmembrane domains and one putative C-terminal phosphoinositide-binding domain. Although PIRT is expressed in peripheral nervous system, with highest levels in dorsal root ganglion and trigeminal neurons, and lowest levels in sympathetic and enteric neurons, it is not expressed in spinal cord. PIRT is a required component of the VR1 complex, which positively regulates VR1, a sensor of both noxious heat and capsaicin. Correspondingly, PIRT knockout results in impaired responses to noxious heat and capsaicin exposure, while VR1 remains unaltered. The gene that encodes PIRT maps to human chromosome 17p13.1.

Mutations of ankyrin genes lead to severe genetic diseases such as fatal cardiac arrhythmias and hereditary spherocytosis. ANKLE2 (ankyrin repeat and LEM domain containing 2), also known as LEMD7, is a 938 amino acid single-pass membrane protein containing an ANK repeat and a LEM domain. Exsiting as two isoforms produced by alternative splicing events, the gene encoding ANKLE2 maps to human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome. Chromosome 12 is associated with a variety of diseases and afflictions, including hypochondrogenesis, achondrogenesis, Kniest dysplasia, Noonan syndrome and trisomy 12p, which causes facial developmental defects and seizure disorders.
