PKMYT1 is a member of the serine/threonine protein kinase family. It preferentially phosphorylates and inactivates cell division cycle 2 protein (cdc2), and thus acts as a negative regulator of entry into mitosis (G2 to M transition). It mediates phosphorylation of cdc2 predominantly on 'Thr-14' and is also involved in Golgi fragmentation. It may be involved in phosphorylation of cdc2 on 'Tyr-15' to a lesser degree, however tyrosine kinase activity is unclear and may be indirect. It may be a downstream target of Notch signaling pathway during eye development. PKMYT1 is negatively regulated by hyperphosphorylation during mitosis.

Kindlin-3 is a 667 amino acid protein that localizes to both the cell membrane and the cytoplasm and contains one PH domain and one FERM domain. Expressed at high levels in lymph node tissue and at lower levels in spleen, thymus, stomach, placenta, lung, testis and small intestine, Kindlin-3 is thought to be involved in cell adhesion events and may play a role in apoptosis. Kindlin-3 is overexpressed in B-cell malignancies, suggesting that, via its ability to affect cell adhesion, Kindlin-3 may participate in tumor transformation and metastasis. Two isforms of Kindlin-3, designated short and long, exist due to alternative splicing events.

In addition, both proteins can inhibit the transcription of cell cycle genes containing E2F binding sites in their promoters. Due to the sequence and biochemical similarities with the RB1 protein, it is thought that the protein encoded by this gene may also be a tumor suppressor. Two transcript variants encoding different isoforms have been found for this gene.

The ubiquitin (Ub) pathway involves three sequential enzymatic steps that facilitate the conjugation of Ub and Ub-like molecules to specific protein substrates. The first step requires the ATP-dependent activation of the Ub C-terminus and the assembly of multi-Ub chains by the Ub-activating enzyme known as the E1 component. The Ub chain is then conjugated to the Ub-conjugating enzyme (E2) to generate an intermediate Ub-E2 complex. The Ub-ligase (E3) then catalyzes the transfer of Ub from E2 to the appropriate protein substrate. A wide range of enzymes facilitate in the proteolytic Ub pathway, including monoclonal nonspecific suppressor factor-beta (MNSF-beta), a subunit of MNSF, which is a lymphokine product of a murine T cell hybrid-oma that restricts the production of LPS-induced immunoglobulin secreting cells in an antigen-nonspecific manner. MNSF-beta is a ubiquitin-like fusion protein consisting of the ribosomal protein S30 and a protein that shares 36% sequence identity with ubiquitin. This ubiquitin-like segment (Ubi-L) can be cleaved from MNSF-beta in the cytosol.
