Description:
CD31 (PECAM-1) is a transmembrane glycoprotein member of the immunoglobulin supergene family of adhesion molecules. CD31 is expressed by stem cells of the hematopoietic system and is primarily used to identify and concentrate these cells for experimental studies as well as for bone marrow transplantation. Anti-CD31 has shown to be highly specific and sensitive for vascular endothelial cells. Staining of nonvascular tumors (excluding hematopoietic neoplasms) is rare. CD31 MAb reacts with normal, benign, and malignant endothelial cells which make up blood vessel lining. The level of CD31 expression can help to determine the degree of tumor angiogenesis, and a high level of CD31 expression may imply a rapidly growing tumor and potentially a predictor of tumor recurrence.

Description:
Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. This MAb selectively interferes with lymphocyte binding to lymph node, mucosal and synovial endothelium. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. CD44 immunostaining is commonly used for the discrimination of urothelial transitional cell carcinoma in-situ from non-neoplastic changes in the urothelium.

Description:
Expression of DOG-1 protein is elevated in the gastrointestinal stromal tumors (GISTs), c-kit signaling-driven mesenchymal tumors of the GI tract. DOG-1 is rarely expressed in other soft tissue tumors, which, due to appearance, may be difficult to diagnose. Immunoreactivity for DOG-1 has been reported in 97.8 percent of scorable GISTs, including all c-kit negative GISTs. Overexpression of DOG-1 has been suggested to aid in the identification of GISTs, including Platelet-Derived Growth Factor Receptor Alpha mutants that fail to express c-kit antigen. The overall sensitivity of DOG1 and c-kit in GISTs is nearly identical: 94.4% vs. 94.7%.

Description:
This antibody recognizes an intra-cytoplasmic antigen, which shows a very high degree of specificity for plasma cells. This antigen is present in normal as well as neoplastic plasma cells. Plasma cells, which are large lymphocytes derived from an antigen-specific B cell, secrete antibodies and are responsible for humoral immunity. Plasma cells differentiate from B cells upon stimulation by CD4 lymphocytes. The B cell acts as an antigen-presenting cell (APC), consuming an offending pathogen, which is taken up by the B cell by phagocytosis and broken down within proteosomes. Plasma cells contain basophilic cytoplasm; their nucleus contains heterochromatin organized in a characteristic cartwheel arrangement. This MAb superbly recognizes normal and neoplastic plasma cells in routine formalin-fixed, paraffin-embedded tissue sections. It is of potential value in identifying myeloma or plasmacytoma in bone marrow or other tissues. It also helps differentiate lympho-plasmacytoid lymphoma from lymphocytic and follicular lymphoma. Note that this MAb is not suitable for staining frozen tissues.

Description:
This antibody recognizes a protein of 70 kDa, identified as prolactin receptor. Prolactin is a pituitary hormone involved in the stimulation of milk production, salt and water regulation, growth, development and reproduction. The initial step in its action is the binding to a specific membrane receptor (prolactin receptor), which belongs to the superfamily of class 1 cytokine receptors. The function of the prolactin receptor is mediated, at least in part, by two families of signaling molecules: Janus kinases and signal transducers and activators of transcription.

Description:
Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained.Immunohistochemical staining with this MAb is indistinguishable from that obtained with monoclonal antibody 5D3.

Description:
This MAb is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells. This monoclonal antibody is part of a new panel of reagents, which recognizes subcellular organelles or compartments of human cells. These markers may be useful in identification of these organelles in cells, tissues, and biochemical preparations. MAb NM95 recognizes an antigen associated with the nucleoli in human cells. It can be used to stain the nucleoli in cell or tissue preparations and can be used as a marker of the nucleoli in subcellular fractions. It produces a speckled pattern in the nuclei of cells of normal and malignant cells and may be used to stain the nucleoli of cells in fixed or frozen tissue sections. It can be used with paraformaldehyde fixed frozen tissue or cell preparations and formalin fixed, paraffin-embedded tissue sections.

Description:
This MAb recognizes full-length MUC1 in a glycosylation-independent manner and can bind to the fully glycosylated protein. The dominant epitope of this MAb is APDTR in the VNTR region. It reacts with the core peptide of the MUC1 protein, which is a member of a family of mucin glycoproteins that are characterized by high carbohydrate content, O-linked oligosaccharides, high molecular weight (>200 kDa) and an amino acid composition rich in serine, threonine, proline and glycine. The core protein contains a domain of 20 amino-acid tandem repeats that functions as multiple epitopes for the MAb. Incomplete glycosylation of some tumor-associated mucins may lead to variable unmasking of the multiple peptide epitopes leading to the observed differences in staining intensity between normal and malignant tissues. This MAb reacts with both normal and malignant epithelia of various tissues including breast and colon.

Description:
This antibody recognizes actin of skeletal, cardiac, and smooth muscle cells. It is not reactive with other mesenchymal cells except for myoepithelium. Actin can be resolved on the basis of its isoelectric points into three distinctive components: alpha, beta, and gamma in order of increasing isoelectric point. Anti-muscle specific actin recognizes alpha and gamma isotypes of all muscle groups. Non-muscle cells such as vascular endothelial cells and connective tissues are non-reactive. Also, neoplastic cells of non-muscle-derived tissue such as carcinomas, melanomas, and lymphomas are negative. It stains tumors of smooth muscle (leiomyomas and leiomyosarcomas) as well as skeletal muscle (rhabdomyomas and rhabdomyosarcomas).

Description:
This MAb reacts with a 200 kDa protein, identified as heavy sub-unit of neurofilaments (NF-H). Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. Carcinoids, neuroendocrine carcinomas of the skin, and oat cell carcinomas of the lung also express neurofilament.

Description:
MAb VU-2G7 reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. The dominant epitope of VU-2G7 has not been established with epitope fingerprinting. The extracellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion.

Description:
Recognizes a phosphor-protein of 45 kDa, identified as MyoD1. This MAb does not cross react with myogenin, Myf5, or Myf6. Antibody to MyoD1 labels the nuclei of myoblasts in developing muscle tissues. MyoD1 is not detected in normal adult tissue, but is highly expressed in the tumor cell nuclei of rhabdomyosarcomas. Occasionally nuclear expression of MyoD1 is seen in ectomesenchymoma and a subset of Wilm s tumors. Weak cytoplasmic staining is observed in several non-muscle tissues, including glandular epithelium and also in rhabdomyosarcomas, neuroblastomas, Ewing s sarcomas and alveolar soft part sarcomas.
