TIMP3 (tissue inhibitor of metalloproteinases 3), along with family members TIMP1, TIMP2, and TIMP4, are inhibitors of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix (ECM). An imbalance between MMPs and the associated TIMPs may play a significant role in the invasive phenotype of malignant tumors. TIMP s inhibit the proteolytic invasiveness of tumor cells and normal placental trophoblast cells. TIMP-3 may be involved in regulating trophoblastic invasion of the uterus as well as in regulating remodeling of the extracellular matrix during the folding of epithelia, and in the formation, branching and expansion of epithelial tubes.

Description:
Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five non-covalently linked units, but shows a single 17 kDa band following SDS PAGE under non-reducing conditions. TNF alpha is closely related to the 25 kDa protein Tumor Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production. TNF alpha is currently being evaluated in treatment of certain cancers and AIDS Related Complex.

Recognizes a protein of 36 kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.

This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It was reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. Tissues from gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18.

Recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.4). It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. It acts as calcium channel involved in B cell activation and cell cycle progression.

This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk s and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. Loss of p21 expression has been reported in many carcinomas (gastric carcinoma, non-small cell lung carcinoma, thyroid carcinoma). In ELISA, MAb WA-1 is useful either as a solid phase or for detection of p21 protein.

Epstein-Barr virus (EBV), also designated human herpesvirus 4 (HHV-4), is a member of the herpesvirus family and is one of the most common human viruses. EBV infects B cells and, though often asymptomatic, it can cause infectious mononucleosis, a disease characterized by fatigue, fever, sore throat and muscle soreness. The EBV-induced early antigens (Ea) are among several antigen complexes that have been identified in EBV-infected cells. The Ea complex is composed of diffuse (Ea-D) and restricted (Ea-R) components. The activity of Ea-D is suppressed during latent infection. BMRF1, the gene that encodes for Ea-D, is closely associated with the gene encoding for EBV DNA polymerase, and Ea-D is essential for the activity of this polymerase. Ea-D forms a complex with EBV DNase and, together, they may play a role in viral replication.

Somatic neoplasms e.g. breast, gastrointestinal, prostatic, and urinary cancers may also immunoreact with antibodies to PLAP. Anti-PLAP positivity in conjunction with anti-keratin negativity favors seminoma over carcinoma. Germ cell tumors are usually anti-keratin positive, but they regularly fail to stain with anti-EMA, whereas most carcinomas stain with anti-EMA. Anti-PLAP has been useful in the diagnosis of gestational trophoblastic disease.

This MAb is specific to Complement 4d (C4d) and it reacts with the secreted as well as cell-bound C4d.C4d is a degradation product of the activated complement factor C4b. Complement 4b is typically activated by binding of Abs to specific target molecules. Following activation and degradation of the C4 molecule, thio-ester groups are exposed, which allow transient, covalent binding of the degradation product Complement 4d to endothelial cell surfaces and extracellular matrix components of vascular basement membranes near the sites of C4 activation. The presence of C4d in peritubular capillaries is a key indicator for acute humoral (i.e. antibody-mediated) rejection of kidney, heart, pancreas and lung allografts. As an established marker of antibody-mediated acute renal allograft rejection and its proclivity for endothelium, this component can be detected in peritubular capillaries in chronic renal allograft rejection as well as hyperacute rejection, acute vascular rejection, acute cellular rejection, and borderline rejection. It has been shown to be a significant predictor of transplant kidney graft survival. Anti-C4d, combined with anti-C3d, can be utilized as a tool for diagnosis of allograft rejection that may warrant a prompt and aggressive anti-rejection treatment.

This antibody recognizes an intra-cytoplasmic antigen, which shows a very high degree of specificity for plasma cells. This antigen is present in normal as well as neoplastic plasma cells. Plasma cells, which are large lymphocytes derived from an antigen-specific B cell, secrete antibodies and are responsible for humoral immunity. Plasma cells differentiate from B cells upon stimulation by CD4 lymphocytes. The B cell acts as an antigen-presenting cell (APC), consuming an offending pathogen, which is taken up by the B cell by phagocytosis and broken down within proteosomes. Plasma cells contain basophilic cytoplasm; their nucleus contains heterochromatin organized in a characteristic cartwheel arrangement. This MAb superbly recognizes normal and neoplastic plasma cells in routine formalin-fixed, paraffin-embedded tissue sections. It is of potential value in identifying myeloma or plasmacytoma in bone marrow or other tissues. It also helps differentiate lympho-plasmacytoid lymphoma from lymphocytic and follicular lymphoma. Note that this MAb is not suitable for staining frozen tissues.
