Description:
Recognizes a 53 kDa protein, which is identified as p53 suppressor gene product. It reacts with the mutant as well as the wild form of p53. Its epitope maps within the N-terminus (aa 37-45) of p53. Monoclonal antibody PAb1801 does not block the binding of DO-7 MAb to p53 in an ELISA test. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Anti-p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intratubular germ cell neoplasia. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.

Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.

Recognizes a protein of 95 kDa, identified as CD19. CD19 is a transmembrane glycoprotein that contains two extracellular immunoglobulin-like domains. CD19 is present in both benign and malignant B-cells and is considered to be the most reliable surface marker of this lineage over a wide range of maturational stages. In normal lymphoid tissue, CD19 is observed in germinal centers, in mantle zone cells, and in scattered cells of the inter-follicular areas. Anti-CD19 exhibits an overall immunoreactivity pattern similar to those of the antibodies against CD20 and CD22. However, in contrast to CD20, expression of CD19 is continuous throughout B-cell development and through terminal differentiation of B-cells into plasma cells. Anti-CD19 positivity is seen in the vast majority of B-cell neoplasms commonly at a lower intensity than normal B-cell counterparts. Plasma cell neoplasms are nearly always negative, as are T-cell neoplasms.

This MAb recognizes a protein of 145 kDa, identified as CD117/p145kit. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma. However, anti-CD117 has been particularly useful in differentiating gastrointestinal stromal tumors from Kaposi's sarcoma, tumors of smooth muscle origin, fibromatosis, and neural tumors of the GI tract. Anti-CD117 is also useful in recognizing myeloblasts in bone marrow biopsy and clot section.

CD11a complex with the 2 subunit of the integrin family, CD18, to form the cell surface heterodimer, LFA-1 or CD11a /C18 (aLbL). LFA-1 is expressed on all leukocytes including lymphocytes, monocytes, and granulocytes. It is involved in leukocyte adhesion to its ligands including intercellular adhesion molecule-1 (ICAM-1 or CD54), ICAM-2 (CD102), ICAM-3 (CD50) and Telencephalin (TLN) and play a role in most immune/inflammatory responses.

Recognizes a protein of ~76 kDa, which is identified as Nucleolin (NCL). It is the major nucleolar phosphoprotein of growing eukaryotic cells. NCL is located mainly in dense fibrillar regions of the nucleolus. It is found associated with intranucleolar chromatin and pre-ribosomal particles. Human NCL gene consists of 14 exons with 13 introns and spans approximately 11kb. It induces chromatin decondensation by binding to histone H1. It is thought to play a role in pre-rRNA transcription and ribosome assembly.This MAb can be used to stain the nucleoli in cell or tissue preparations and can be used as a marker of the nucleoli in subcellular fractions. It produces a speckled pattern in the nuclei of cells of normal and malignant cells and may be used to stain the nucleoli of cells in fixed or frozen tissue sections. This antibody recognizes a protein of ~90 kDa, which is identified as Adenosine Monophosphate Deaminase, isoform E (AMPD3). It has 767 amino acids and is assigned an EC 3.5.4.6. It is a highly regulated enzyme that catalyzes the hydrolytic deamination of adenosine monophosphate to inosine monophosphate, a branch point in the adenylate catabolic pathway. AMPD3 gene encodes the erythrocyte (E) isoforms, whereas other family members encode isoforms that predominate in muscle (M) and liver (L) cells. This MAb shows reactivity with cells of the erythroid lineage at all stages of maturation in the peripheral blood, bone marrow, and fetal liver. Non-erythroid lineages are negative by flow cytometry. This MAb is useful in the diagnosis of erythroleukemia, identification of bone marrow erythroid precursors, gating erythroid nucleated precursor cells from malignant cells in bone marrow specimens.

This antibody recognizes a protein of 25-26 kDa, identified as the bcl-2 α oncoprotein. It shows no cross-reaction with Bcl-x or Bax protein. Expression of bcl-2 α oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of bcl-2 α protein, whereas the normal or hyperplastic germinal centers are negative. Consequently, this antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express bcl-2 protein and the small number in which the neoplastic cells are bcl-2 negative.

Description:
UACA (Uveal Autoantigen with Coiled-coil domains and Ankyrin repeats) is a 1,416 amino acid nuclear membrane protein. It was originally identified as an autoantigen in patients with panuveitis, a characteristic of Vogt-Koyanagi-Harada disease, and in patients with Graves' disease. UACA was also later identified as Nucling, an mRNA differentially expressed in F9 embryonal carcinoma cells during cardiac muscle differentiation. UACA appears to function as a pro-apoptotic protein that recruits the apaf-1-pro-caspase-9 complex for the induction of apoptosis to mediate the cell-death pathway.
