The protein encoded by this gene belongs to the ferlin family and is a skeletal muscle protein found associated with the sarcolemma. It is involved in muscle contraction and contains C2 domains that play a role in calcium-mediated membrane fusion events, suggesting that it may be involved in membrane regeneration and repair. In addition, the protein encoded by this gene binds caveolin-3, a skeletal muscle membrane protein which is important in the formation of caveolae. Specific mutations in this gene have been shown to cause autosomal recessive limb girdle muscular dystrophy type 2B (LGMD2B) as well as Miyoshi myopathy. Functions in the early steps of protein synthesis by forming a ternary complex with GTP and initiator tRNA. This complex binds to a 4S ribosomal subunit, followed by mRNA binding to form a 43S preinitiation complex. Junction of the 6S ribosomal subunit to form the 8S initiation complex is preceded by hydrolysis of the GTP bound to eIF-2 and release of an eIF-2-GDP binary complex. In order for eIF-2 to recycle and catalyze another round of initiation, the GDP bound to eIF-2 must exchange with GTP by way of a reaction catalyzed by eIF-2B.

The isthmic organizer signals at the mid/hindbrain boundary (MHB) regulate the development and differentiation of the vertebrate caudal midbrain and the anterior hindbrain. The MHB forms at the boundary of expression between homeobox genes Gbx2 and Otx2. Gbx2 and Otx2 play distinct, essential roles in MHB positioning and development. During development, the GBX2 gene is expressed in the anterior hindbrain. Specifically, Gbx2 negatively regulates Otx2 expression along the anterior-posterior axis; Gbx2(-) mutants demonstrate an expanded Otx2 domain. During development, the GBX2 gene is expressed in the anterior hindbrain. Gbx2 is expressed in the adult brain, spleen and female genital tract. The GBX2 gene is over-expressed in human prostate cancer cell lines (TSU-prl, PC3, DU145 and LNCaP). Furthermore, downregulation of Gbx2 expression restricts tumorigenicity in human prostate cancer cell lines, which suggests that Gbx2 expression may be required for growth of malignant prostate cells.

The Trk family of nerve growth factor receptors includes Trk A(also referfed to as Trk A gp140),Trk B and Trk C. The prototype member of this gene family, Trk A, encodes a 140 kDa cell surface receptor , gp140, the expression of which is restricted in vivo to neurons of the sensory spinal and cranial gangliaof neurocrest origin. Nerve growth factor (NGF) stimulates tyrosine phosphorylation of Trk gp 140 in neural cell lines and in embryonic dorsal root ganglia. By comparison, BDNF and to a lesser extent, NT-3, but not NGF, can induce tyrosine phophorylayion of Trk B gp 145. The third member of the Trk receptor family, Trk C incodes a 140 kDa protein, Trk C gp140, that is preferentially expressed in brain tissue and primarily functions as a receptor for NT-3.An additional component of the Trk receptor complex, NGFR p175, binds to neurotrophic factors with low affinity but is required for efficient signaling. NGFR p175 accelerates Trk activation and may recruit downstream dffector molecules to the ligand-bound receptor complex.

HS1BP3 is a 392 amino acid protein that contains one PX (phox homology) domain, a leucine zipper, immunoreceptor tyrosine-based inhibitory motif-like motifs and multiple proline-rich regions. Expressed primarily in brain, HS1BP3 is encoded by a gene mapping to human chromosome 2p24.1. The gene encoding HS1BP3 is frequently mutated in familial essential tremor, a disorder characterised by kinetic tremor the the hands, voice or head, though there is no correlation to Parkinson disease. HS1BP3 interacts with HAX-1 SH3 domain, and may also play a role in the regulation of catecholamine and serotonin metabolism. Acting as a regulator of IL-2 Signalling, HS1BP3 is likely involved in lymphocyte activation.

Phelan-McDermid syndrome, Neurofibromatosis type 2 and autism are associated with chromosome 22. A schizophrenia susceptibility locus has been identified on chromosome 22 and studies show that 22q11 deletion symptoms include a high incidence of schizophrenia. Translocations between chromosomes 9 and 22 may lead to the formation of the Philadelphia Chromosome and the subsequent production of the novel fusion protein, BCR-Abl, a potent cell proliferation activator found in several types of leukemia. The C22orf15 gene product has been provisionally designated C22orf15 pending further characterization.

Myotrophin (V-1 protein) is a ubiquitously expressed cytoplasmic protein that can translocate to the nucleus during sustained NFkB activation. The gene encoding for this protein localizes to chromosome 7q33. Myotrophin may be involved in cerebellar morphogenesis and contains an acetylated N-terminus and 2.5 internal 33 amino acid ankyrin repeats. It is important in the differentiation of cerebellar neurons, particularly of granule cells. The 117 amino acid protein has been associated with, and able to induce, cardiac hypertrophy. Myotrophin increases protooncogene, ANF and Beta-Myosin heavy chain transcript levels. Myotrophin is upregulated when myocytes undergo cyclic stretch or are treated with tumor necrosis factor Alpha (TNF Alpha) or interleukin-1Beta. Highest levels of Myotrophin are detected in brain and lowest levels in skeletal muscle.
