Description:   SH2D1A, also SH2 domain protein 1A, SAP and CD150/SLAM (Signalling lymphocyte activation molecule)-associated protein, influences Signalling pathways involving SLAM molecules at the interface between T and B cells. SH2D1A modulates SLAM by blocking the recruitment of tyrosine phosphatase SHP2 to the phosphorylated cytoplasmic domain of SLAM. SLAM activation mediates expansion of activated T cells during immune responses, induces production of interferons and changes the functional profile of subsets of T cells. SH2D1A is a hydrophilic, 128 amino acid protein that is 96% homologous to the mouse protein in both SH2 and tail domains. SH2D1A is present in all major subsets of T cells, including CD⁴⁺, CD45RO⁺, CD45RA⁺ and CD⁸⁺, but not in B cells. SH2D1A can interact via an SH2 domain with a motif (TIYXXV) present in the cytoplasmic tail of cell-surface receptors SLAM (CD150), CD84, CD229 (LY9) and CD244 (2B4).

Description:   MARCH5 is a member of the E3 ubiquitin-protein ligase family. This mitochonrial E3 ubiquitin-protein ligase plays an importnat role in controlling the morphology of the mitochondria. E3 ubiquitin ligases accept ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfer the ubiquitin to targeted substrates. MARCH5 has been shown to interact with MTFN2 and ubiquitinated forms of DRP1.

Description:   Might act as an E3 ubiquitin-protein ligase which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes and then transfers it to substrates, which could be nuclear proteins. Could play a role as a coactivator for androgen- and, to a lesser extent, progesterone-dependent transcription.

Description:   ANKRD17 is a 2,603 amino acid protein that contains 25 ankyrin repeats and one KH domain. ANKRD17 is expressed in bone marrow and is thought to be involved in liver development. ANKRD17 localizes to the cytoplasm and the nucleus. ANKRD17 exists as five alternatively spliced isoforms that are encoded by a gene which maps to human chromosome 4. Representing approximately 6% of the human genome, chromosome 4 contains nearly 900 genes. Notably, the Huntingtin gene, which is found to encode an expanded glutamine tract in cases of Huntington's disease, is on chromosome 4. FGFR-3 is also encoded on chromosome 4 and has been associated with thanatophoric dwarfism, achondroplasia, Muenke syndrome and bladder cancer. Chromosome 4 is also tied to Ellis-van Creveld syndrome, methylmalonic acidemia and polycystic kidney disease. Chromosome 4 reportedly contains the largest gene deserts (regions of the genome with no protein encoding genes) and has one of the two lowest recombination frequencies of the human chromosomes.

Description:   14-3-3 proteins regulate many cellular processes relevant to cancer biology, notably apoptosis, mitogenic signaling and cell-cycle checkpoints. Seven isoforms comprise this family of signaling intermediates, denoted 14-3-3 b, g, e, z, h, q and s. 14-3-3 proteins form dimers that present two binding sites for ligand proteins, thereby bringing together two proteins that may not otherwise associate. These ligands largely share a 14-3-3 consensus binding motif and exhibit serine/threonine phosphorylation. 14-3-3 proteins function in broad regulation of these ligand proteins; by cytoplasmic sequestration, occupation of interaction domains and import/export sequences, prevention of degradation, activation/repression of enzymatic activity, and facilitation of protein modification. Loss of expression contributes to a vast array of pathogenic cellular activities.

Description:   14-3-3 proteins regulate many cellular processes relevant to cancer biology, notably apoptosis, mitogenic signaling and cell-cycle checkpoints. Seven isoforms, denoted 14-3-3 b, g, e, z, h, q and s, comprise this family of signaling intermediates. 14-3-3 s, also known as SFN, stratifin, HME1 or YWHAS, is a secreted adaptor protein that is involved in regulating both general and specific signaling pathways. Expressed predominately in stratified squamous keratinising epithelium, 14-3-3 s is able to bind and modify the activity of a large number of proteins, such as KRT17 (Keratin 17), through recognition of a phosphothreonine or phosphoserine motif. When bound to Keratin 17, for example, 14-3-3 s acts to stimulate the Akt/mTOR signaling pathway by upregulating protein synthesis and cell growth. 14-3-3 s also functions to positively mediate IGF-I-induced cell cycle progression and can bind to a variety of translation initiation factors, thus controlling mitotic translation. In response to tumor growth, 14-3-3 s positively regulates the tumor suppressor p53 and increases the rate of p53-regulated inhibition of G2/M cell cycle progression. Multiple isoforms of 14-3-3 s exist due to alternative splicing events.

Description:   Making up nearly 6% of the human genome, chromosome 6 contains around 1200 genes within 170 million base pairs of sequence. Deletion of a portion of the q arm of chromosome 6 is associated with early onset intestinal cancer suggesting the presence of a cancer susceptibility locus. Porphyria cutanea tarda is associated with chromosome 6 through the HFE gene which, when mutated, predisposes an individual to developing this porphyria. Notably, the PARK2 gene, which is associated with Parkinson's disease, and the genes encoding the major histocompatiblity complex proteins, which are key molecular components of the immune system and determine predisposition to rheumatic diseases, are also located on chromosome 6. Stickler syndrome, 21-hydroxylase deficiency and maple syrup urine disease are also associated with genes on chromosome 6. A bipolar disorder susceptibility locus has been identified on the q arm of chromosome 6. The C6orf146 gene product has been provisionally designated C6orf146 pending further characterisation.
