Description:
This gene encodes an accessory subunit of the mitochondrial membrane respiratory chain NADH dehydrogenase (Complex I), or NADH:ubiquinone oxidoreductase, the first multi-subunit enzyme complex of the mitochondrial respiratory chain. Complex I plays a vital role in cellular ATP production, the primary source of energy for many crucial processes in living cells. It removes electrons from NADH and passes them by a series of different protein-coupled redox centers to the electron acceptor ubiquinone. In well-coupled mitochondria, the electron flux leads to ATP generation via the building of a proton gradient across the inner membrane. Complex I is composed of at least 41 subunits, of which 7 are encoded by the mitochondrial genome and the remainder by nuclear genes. [provided by RefSeq, Jul 2008].

The NADH-ubiquinone oxidoreductase complex (complex I) of the mitochondrial respiratory chain catalyzes the transfer of electrons from NADH to ubiquinone, and consists of at least 43 subunits. The complex is located in the inner mitochondrial membrane. This gene encodes the 24 kDa subunit of complex I, and is involved in electron transfer. Mutations in this gene are implicated in Parkinson's disease, bipolar disorder, schizophrenia, and have been found in one case of early onset hypertrophic cardiomyopathy and encephalopathy. ERCs (ERC1 and ERC2) also referred to as ELKS and CAST are related proteins which share an identical C-terminal sequence. They interact with the conserved RIM PDZ domain via an unusual PDZ binding motif. ERC2/CAST 1 is only expressed as a single RIM binding variant. All ERCs have been shown to interact with Rab6, a protein involved in membrane trafficking at the Golgi complex. The function of these proteins has not been determined yet. They may link Rab6 mediated non-neuronal membrane traffic at the Golgi complex to neuronal membrane traffic at the active zone executed via RIMs.

Expressed as two isoforms produced by alternative splicing, TSPEAR contains one Thrombospondin N-terminal domain and seven EAR (epilepsy-associated repeat) domains. EAR domains are found in several proteins, including TSPEAR, encoded by genes that map within chromosome regions associated with seizure disorders. It is thought that the EAR domain plays a role in the pathogenesis of epilepsy by either binding to an unknown epileptic ligand or interfering with axon synaptogenesis.

BCL2 is an integral outer mitochondrial membrane protein that blocks the apoptotic death of some cells such as lymphocytes. Constitutive expression of BCL2, such as in the case of translocation of BCL2 to Ig heavy chain locus, is thought to be the cause of follicular lymphoma. Two transcript variants (alpha and beta) produced by alternate splicing, differ in their C-terminal ends. BCL2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. It appears to function in a feedback loop system with caspases. BCL2 inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF1). It can form homodimers, and heterodimers with BAX, BAD, BAK and BclX(L). Heterodimerization with BAX requires intact BH1 and BH2 domains, and is necessary for anti-apoptotic activity. Also interacts with APAF1, RAF1, TP53BP2, BBC3, BCL2L1 and BNIPL.
