Ephrin B proteins are thought to play key roles in cellular functions as diverse as neuronal migration and blood vessel development. Ephrin B molecules expressed at the membrane surface bind to the Ephrin B family receptors on target cells during cell to cell contact. This interaction leads to cell signaling in the target cell but also generates a reverse signal in the cell expressing Ephrin B on its surface. This reverse signaling event is thought to be critical for vessel maturation and neuronal development. Importantly, tyrosine phosphorylation of Ephrin B is thought to be a critical component of this reverse signaling event. Recent work demonstrated that Tyr331 of Ephrin B was phosphorylated in HEK293 cells after stimulation by the soluble Ephrin B2 receptor tyrosine kinase.

Description:
ALX1 is a 326 amino acid protein specific to cervix and cartilage tissues. As well as having a homeobox domain, ALX1 also contains an OAR domain, which has been suggested to be important for DNA binding or protein-protein interactions and transactivation. First characterized from a rat chondrosarcoma tumor cell line, ALX1 is a homeobox transcription factor that regulates downstream target genes and has specifically shown to act as a transcriptional repressor for rat prolactin in vivo. Homozygous ALX1 deficient mice are born with acrania and meroanencephaly, suggesting ALX1 function in the development of the neural tube. It has also been suggested that ALX1 cooperates with ALX4 in limb development and craniofacial bone formation.

The Olfactomedin family comprises a diverse group of secreted glycoproteins, which includes OLFM1 (Noelin-1), OLFM2 (Noelin-2), OLFM3 (Noelin-3), OLFM4 (Noelin-4), tiarin, pancortin, gliomedin and mycocilin. These proteins are implicated in the development of the nervous system. Specifically, OLFM1 and OLFM2 expression is observed in the neural plate and neural crest, as well as in the cranial ganglia in mouse at E8-10, and later in brain tissue and in the zone of polarizing activity in the limb. Overexpression of OLFM1 causes an excess of neural crest emigrations and prolonged neural crest production. OLFM2 participates in the regulation of the development of the anterior nervous system. An Arg144Gln mutation in OLFM2 has been implicated as a possible cause for open-angle glaucoma (OAG).
