GHRH-R is a seven transmembrane domain protein that localises to the somatotroph of the pituitary. GHRH-R plays an important role in growth and acts as a high-affinity receptor for GHRH. Binding of GHRH leads to the coupling of GHRH-R to G-protein which stimulates increased adenylyl cyclase activity and the accumulation of cAMP leading to the synthesis and release of growth hormone and somatotroph proliferation. In addition, this signalling pathway may have direct action in fetal/placental development, reproduction and immune function. GHRH and GHRH-R may also play a role in the regulation of non-rapid eye movement sleep (NREMS). The expression of GHRH-R is dependent on the presence of the POU domain factor Pit-1. Mutations in the gene encoding this protein can result in isolated growth hormone deficiency (IGHD), also known as Dwarfism of Sindh, and anterior pituitary hypoplasia (APH).

GIMAP3p is a murine mitochondrial membrane protein belonging to the GTP-binding superfamily and to the immuno-associated nucleotide (IAN) subfamily of nucleotide-binding proteins. The IAN/GIMAP family consists of GTP-binding proteins that share a unique primary structure and whose expression is finely regulated by T-cell receptor signals. Recent studies have shown that IAN/GIMAP family proteins crucially regulate the survival of T cells during development, selection and homeostasis, and are possibly linked to the onset of T-lymphopenia, leukemia and autoimmunity. IAN/GIMAP family proteins might also take part in mitochondrial regulation of lymphocyte apoptosis by interacting with Bcl-2 family proteins. The human GIMAP (GTPase of the immunity-associated protein) gene family includes seven functional members residing on (human) chromosome 7q36.1 and one pseudogene (hGIMAP3).

ECAT1 (ES cell-associated transcript 1 protein) is a 217 amino acid protein that belongs to the KHDC1 family. The ECAT1 protein contains an atypical KH domain with amino acid changes at critical sites, suggesting that it may not bind RNA. Expression of ECAT1 appears to be maximal in germinal vesicle oocytes, it tails off through metaphase II oocytes and is undetectable following the completion of the oocyte to embryo transition. Specifically expressed in the oocytes, recent studies suggest that ECAT1 may function as a regulator of genomic imprinting in the oocyte. Defects in ECAT1 are the cause of hydatidiform mole recurrent type 2 (HYDM2), a disorder characterized by excessive trophoblast development that produces a growing mass of tissue inside the uterus at the beginning of a pregnancy. HYDM2 leads to abnormal pregnancies with no embryo, and cystic degeneration of the chorionic villi.

The 53-6.7 antibody reacts with the 32-34 kDa alpha subunit of mouse CD8, known as CD8a or CD8 alpha. CD8a can form a homodimer (CD8 alpha-alpha), but is more commonly expressed as a heterodimer with a second chain known as CD8b or CD8 beta. CD8 acts as a co-receptor in antigen recognition and subsequent T cell activation that is initiated upon binding of the T cell receptor (TCR) to antigen-bearing MHC Class I molecules. The cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells (CTLs) play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites.

There are three SorCS genes that have diverse, partially overlapping functions in the central nervous system. In the developing and mature central nervous system, SorCS1, SorCS2 and SorCS3 genes are expressed in a combinatorial, non-overlapping pattern. SorCS proteins show homology to the mosaic receptor SorLA and the neurotensin receptor sortilin, based on a common VPS10 domain, which is the hallmark of the SorCS receptor family. SorCS2 (sortilin-related VPS10 domain containing receptor 2) is a 1,150 amino acid single-pass type I membrane protein that is highly expressed in brain and kidney. Containing six BNR repeats and a single PKD domain, SorCS2 is encoded by a gene that maps to human chromosome 4, which encodes nearly 6% of the human genome and has the largest gene deserts (regions of the genome with no protein encoding genes) of all of the human chromosomes.

Thrombopoietin (TPO or THPO), also known as c-Mpl ligand (c-Mpl L), is a cytokine that plays a central role in megakaryopoiesis by influencing the development and maturation of megakaryocytes and platelet production from hematopoietic stem cells. TPO exerts its biological effects through the TPO receptor, c-Mpl. c-Mpl is a member of the cytokine receptor superfamily. Expression of c-Mpl is restricted to hematopoietic tissues and cells, such as bone marrow, spleen, fetal liver and CD34+ cells. Stimulation of c-Mpl with TPO results in the activation of the Janus tyrosine kinase family members, Tyk 2 and JAK2, which in turn phosphorylate Stat5 and Stat3, causing their nuclear translocation and the transcription of Stat responsive genes. Muta-tions in c-Mpl have been implicated as the cause of certain human disorders, including congenital amegakaryocytic thrombocytopenia (CAMT) and thrombocytopenia with absent radii (TAR) syndrome.

Semaphorins are a family of cell surface and secreted proteins involved in neural development that are conserved from insects to humans. Members of this family are approximately 750 amino acids in length (including signal sequences) and are defined by a conserved extracellular “semaphorin†domain of approximately 500 amino acids containing 14-16 cysteines, blocks of conserved sequences and no obvious repeats. The transmembrane semaphorins are characterized by an additional 80 amino acid transmembrane domain and an 80-110 amino acid cytoplasmic domain. SEMA6C, also known as SEMA Y, is a transmembrane protein expressed in fetal brain and adult skeletal muscle. Three isoforms of this semaphorin exist due to alternative splicing: SEMA6C 1, SEMA6C 2 and SEMA6C 3. The extracellular domain of SEMA6C induces growth cone collapse of dorsal root ganglion and plays a role in generation or stability of entorhino-hippocampal synapses.

Containing a basic helix-loop-helix (bHLH) domain and an Orange domain, HESL self-associates and interacts with HES5 and HRT2. HESL is considered a transcriptional repressor that binds to the canonical E box sequence 5'-CACGCG-3'. HESL exists as two isoforms produced by alternative splicing events.

FANK1 (fibronectin type III and ankyrin repeat domains 1), also known as HSD13, is a 345 amino acid nuclear and cytoplasmic testis-specific protein found primarily in pachytene spermatocytes and round spermatids. Containing six ANK repeats and a single fibronectin type-III domain, FANK1 undergoes alternative splicing events to form three isoforms. Possessing DNA binding activity, FANK1 is suggested to act as a transcription factor and may regulate gene expression during spermatogenesis. The gene encoding FANK1 maps to human chromosome 10, which contains over 800 genes and 135 million nucleotides, making up nearly 4.5% of the human genome. PTEN is an important tumor suppressor gene located on chromosome 10 and, when defective, causes a genetic predisposition to cancer development known as Cowden syndrome.
