Hydrolyzes lysophospholipids to produce lysophosphatidic acid (LPA) in extracellular fluids. Major substrate is lysophosphatidylcholine. Also can act on sphingosylphosphphorylcholine producing sphingosine-1-phosphate, a modulator of cell motility. Can hydrolyze, in vitro, bis-pNPP, to some extent pNP-TMP, and barely ATP. Involved in several motility-related processes such as angiogenesis and neurite outgrowth. Acts as an angiogenic factor by stimulating migration of smooth muscle cells and microtubule formation. Stimulates migration of melanoma cells, probably via a pertussis toxin-sensitive G protein. May have a role in induction of parturition. Description:
The DAZ (Deleted in AZoospermia) gene family encodes potential RNA binding proteins that are expressed in prenatal and postnatal germ cells of males and females. The protein encoded by this gene is localized to the nucleus and cytoplasm of fetal germ cells and to the cytoplasm of developing oocytes. In the testis, this protein is localized to the nucleus of spermatogonia but relocates to the cytoplasm during meiosis where it persists in spermatids and spermatozoa. Transposition and amplification of this autosomal gene during primate evolution gave rise to the DAZ gene cluster on the Y chromosome. Mutations in this gene have been linked to severe spermatogenic failure and infertility in males. Two transcript variants encoding different isoforms have been found for this gene. The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. RNF215 (ring finger protein 215), is a 377 amino acid multi-pass membrane protein containing one RING-type zinc finger. The gene encoding RNF215 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

The Forkhead-box (FOX) genes comprise a superfamily of at least 43 members that encode proteins which are involved in transcriptional regulation and may be associated with the pathogenesis of various cancers. FOXB1 (forkhead box B1), also known as FKH5 or HFKH-5, and FOXB2 (forkhead box B2) are members of the FOX family and each contain one forkhead DNA-binding domain. Both FOXB1 and FOXB2 localize to the nucleus where they are thought to function as transcription factors that can bind to DNA via their forkhead domains. In mice, defects in the gene encoding FOXB1 are associated with retarded development of the central nervous system (CNS), suggesting that FOXB1 may play a role in CNS organization and function.

Neuronal migration of the central nervous system is a specialized form of cell motility that takes place in the laminar structure of the cortical regions of brain. Astrotactin is a neuronal cell surface protein expressed on postmitotic neuronal precursors in the cerebellum, hippocampus, cerebrum, and olfactory bulb. Astrotactin 2 is a paralog of Astrotactin. Astrotactin mediates neuron-astroglial interactions and is also implicated in synaptic development as well as many other neuronal activities. Astrotactin has three epidermal growth factor repeat domains and two fibronectin type III repeat domains. The human Astrotactin gene shows extensive homology to the mouse Astrotactin gene. Mutations in the Astrotactin gene are linked to neuronal migration defects in both species.

Filensin, also referred to as CP94, CP95, or CP97, is an eye lens fiber, membrane-associated, cytoskeletal intermediate filament (IF) protein that is required for the assembly of beaded filaments, cytoskeletal networks that are necessary for the long-term maintenance of optical clarity. Phakinin copolymerizes with Filensin to make up the filamentous structures present in the beaded filaments. Filensin is also crucial for lens development since it regulates lens fiber cell shape conformation and lens transparency. Filensin contains a C-terminal non-α-helical domain that contributes in several ways to its function. Tight junctions help establish polarity in mammalian epithelia by forming a physical barrier that separates apical and basolateral membranes. Pals-associated tight junction protein (PATJ), the human homolog of Drosophila Discs Lost, is differentially localized in myelinating Schwann cells. PATJ associates with Claudin-1, CRB1 (a transmembrane protein that plays a role in epithelial cell polarity and photoreceptor development), and Pals1 (a Lin-7 associated protein). The PATJ/Pals1/CRB1 complex can form a tripartite tight junction in epithelial cells crucial to their integrity.

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene encodes a replication-independent member of the histone H2A family that is distinct from other members of the family. Studies in mice have shown that this particular histone is required for embryonic development and indicate that lack of functional histone H2A leads to embryonic lethality. Tuberous sclerosis complex (TSC) is an autosomal dominant genetic disorder characterised by mental retardation and the widespread development of distinctive tumors termed hamartomas. Two different genetic loci have been linked to TSC; one of these loci, the tuberous sclerosis-2 gene (TSC2), encodes a protein called tuberin and the other loci, tuberous sclerosis-1 gene (TSC1), encodes a protein called hamartin. Tuberin and hamartin interact with each other forming a cystoplasmic complex. Hamartin interacts with the ezrin-radixin-moesin (ERM) family of actin-binding proteins and inhibition of hamartin activity results in loss of cell adhesion. Hamartin is present in most adult tissues with strong expression in brain, heart, and kidney.
