Description:
Fractalkine (CX3CL1) is a unique chemokine belonging to the CX3C family, and is characterized by a C-X3-C cysteine motif within the chemokine domain, near the amino terminus of the protein (Bazan <i>et al.</i>). The chemokine domain is connected to an extended mucin-like stalk, followed by a transmembrane region, and a C-terminal intracellular domain (Imai <i>et al.</i>; Jones <i>et al.</i>). The protein signals through interaction with a single receptor, CX3CR1, expressed on monocytes, natural killer cells, T cells, microglia, and smooth muscle cells. Fractalkine is upregulated in endothelial cells by inflammatory signals and is synthesized as a membrane-bound molecule that mediates cell migration and adhesion (White and Greaves). Cleavage at the base of the stalk by metalloproteinases generates a soluble chemokine, which functions as a potent chemoattractant of target cells (Garton <i>et al.</i>; Apostolakis and Spandidos). Fractalkine has been implicated in pathology of inflammatory diseases, such as atherosclerosis and other vascular diseases, and has anti-apoptotic functions (White and Greaves).

Description:
This gene encodes a common acute lymphocytic leukemia antigen that is an important cell surface marker in the diagnosis of human acute lymphocytic leukemia (ALL). This protein is present on leukemic cells of pre-B phenotype, which represent 85% of cases of ALL. This protein is not restricted to leukemic cells, however, and is found on a variety of normal tissues. It is a glycoprotein that is particularly abundant in kidney, where it is present on the brush border of proximal tubules and on glomerular epithelium. The protein is a neutral endopeptidase that cleaves peptides at the amino side of hydrophobic residues and inactivates several peptide hormones including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. This gene, which encodes a 100-kD type II transmembrane glycoprotein, exists in a single copy of greater than 45 kb. The 5' untranslated region of this gene is alternatively spliced, resulting in four separate mRNA transcripts. The coding region is not affected by alternative splicing. [provided by RefSeq].

Interleukin-6 receptor subunit beta (IL6ST) is also known as IL-6 receptor subunit beta, IL-6R subunit beta, IL-6R-beta, IL-6RB, Interleukin-6 signal transducer, Membrane glycoprotein 130 (gp130), CD130, Oncostatin-M receptor subunit alpha and Il6st,which is single-pass type I membrane protein. IL6ST /gp130 /CD130 can be found in tissues such as brain, heart, thymus, spleen, kidney, lung and liver and found in all the cell lines tested except BaF-B03.The expression of IL-6ST /gp130 is not restricted to IL6-responsive cells. The receptor systems for IL6, LIF, OSM, CNTF, IL11, CTF1 and BSF3 can utilize gp130 for initiating signal transmission. IL6ST /CD130 can bind to IL6 /IL6R (alpha chain) complex, resulting in the formation of high-affinity IL6 binding sites, and transduce the signal. IL6ST /GP130 does not bind IL6 and may have a role in embryonic development.

CD antigen CD98 is also known as 4F2 cell-surface antigen heavy chain (4F2hc), 4F2 heavy chain antigen, Solute carrier family 3 member 2 (SLC3A2), Lymphocyte activation antigen 4F2 large subunit, is a single-pass type I I membrane protein which belongs to the SLC3A transporter family.. SLC3A2 / CD98 is expressed ubiquitously in all tissues tested with highest levels detected in kidney, placenta and testis and weakest level in thymus. SLC3A2 / CD98 is required for the function of light chain amino-acid transporters and also involved in sodium-independent, high-affinity transport of large neutral amino acids such as phenylalanine, tyrosine, leucine, arginine and tryptophan. CD98 involved in guiding and targeting of LAT1 and LAT2 to the plasma membrane. When associated with SLC7A5 or SLC7A8, CD98 involved in the cellular activity of small molecular weight nitrosothiols, via the stereoselective transport of L-nitrosocysteine (L-CNSO) across the transmembrane. Together with ICAM1, regulates the transport activity LAT2 in polarized intestinal cells, by generating and delivering intracellular signals.

RNAse H2A Antibody: Ribonucleases (RNAses) H are enzymes that hydrolyze the RNA strands of RNA/DNA hybrids. The major role of these enzymes is to remove the RNA strand from the RNA/DNA hybrids that form during DNA replication and repair. RNAse H2 is made up of three subunits; all three are required for RNAse activity. Recent evidence has demonstrated that mutations in RNAse H2A or any of the other subunits result in Aicardi-Goutieres syndrome (AGS), a neurological disorder with similar symptoms to viral brain infections including high levels of IFN-alpha in the cerebral spinal fluid. Similar conditions are observed with mutations in TREX1, a single-stranded DNA exonuclease, suggesting that RNAse H2 and TREX1 may have similar roles, and that mutations in any of these genes lead to an accumulation of intracellular nucleic acids, triggering an inflammatory response through activation of the innate immune system.

The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus belongs to receptor type PTP. This gene is specifically expressed in hematopoietic cells. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling. Mouse interleukin-16(IL-16) is a single chain non-glycosylated polypeptide. IL-16 is widely expressed in human tissues including spleen, thymus, lymph nodes, peripheral leukocytes, bone marrow and cerebellum. IL-16 plays an important role instimulating a migratory response in CD4+ lymphocytes, monocytes, and eosinophils,inducing T-lymphocyte expression of interleukin 2 receptor.It was originally identified as a CD8+ T cell-derived chemoattractant for CD4+ cells. In addition to its chemotactic properties, IL-16 has also been shown to suppress HIV-1 replication in vitro and appears to be involved in transcriptional regulation of SKP2 and is probably part of a transcriptional repression complex on the core promoter of the SKP2 gene. It may act as a scaffold for GABPB1 (the DNA-binding subunit the GABP transcription factor complex) and HDAC3 thus maintaining transcriptional repression and blocking cell cycle progression in resting T-cells.

C-Type Lectin Domain Family 10 Member A (CLEC10A) is a type II transmembrane C-type lectin that is expressed on immature myleloid dendritic cells and alternatively activated (tolerogenic) macrophages. CLEC10A/MGL binds and internalizes molecules with terminal nonsialylated GalNAc carbohydrates such as the Tn carcinoma antigen. CLEC10A/MGL also binds the GP envelope glycoprotein on Marburg and Ebola viruses and enhances viral entry and infectivity. It constitute a unique class of C-type lectins because of their specificity for galactose and its structural homologues. CLEC10A is thought to participate in the recognition of molecules from both altered self and pathogens due to its monosaccharide specificity for Gal and N-acetylgalactosamine (GalNAc). Human and rat carry a single gene for CLEC10A/MGL, while mouse has two closely related MGL1 and MGL2 genes.

Ephrin Type-A Receptor 7 (EPHA7) is a single-pass type I membrane protein which belongs to the Eph family of transmembrane receptor tyrosine kinases. It contains two fibronectin type-III domains, one protein kinase domain and one SAM (sterile alpha motif) domain. EPHA7 is a receptor for members of the ephrin-A family. Eph receptors are largely expressed throughout the ectoderm, mesoderm, and endoderm of vertebrate embryos. EPHA7 functions as a repulsive guidance molecule during the targeting of retinal axons to the superior colliculus and of neocortical axons to the thalamus. EPHA7 is expressed at a substantial level in most human lung cancers. The high expression of EPHA7 protein may participate in the malignancy transformation, invasion progression and metastasis of primary hepatocellular carcinoma. EPHA7 may involve in smoking related lung carcinogenesis.
