ERMAP is a single-pass type one membrane protein that belongs to the immunoglobulin superfamily. Expressed in cord blood, fetal liver and adult bone marrow, ERMAP is thought to function as a cell adhesion molecule in erythroid cells and is responsible for expression of the Scianna/Radin (Sc/Rd) blood group antigen system. The Sc/Rd system is comprised of seven antigens that are present on the surface of red blood cells and have a variety of functions ranging from protein transport to cell adhesion. These seven blood antigens can differ in their expression within a population and may sometimes differ between mother and child. A fetus expressing different blood antigens than its mother may cause the mother to produce against the fetal blood. This condition is known as hemolytic disease of the newborn (HDN) and is characterized by jaundice, anemia and in some cases, infant death.

FXYDs are single-span membrane proteins that share a 35 amino acid signature domain, beginning with the sequence PFXYD and containing seven invariant and six conserved amino acids. Members of the FXYD family include FXYD1 (PLM, phospholemman), FXYD2 (the g subunit of the Na+/K+-ATPase), FXYD3 (Mat8, mammary tumor protein), FXYD4 (CHIF) and FXYD5 (RIC). FXYD6 is expressed in various epithelial cells bordering the endolymph space and in the auditory neurons. CatSpers are vital to sperm motility, fertilization and cAMP-mediated calcium influx in sperm. There are four CatSper proteins in mammalian sperm, namely CatSper (or CatSper1), CatSper2, CatSper3 and CatSper4. The UNC5H family of proteins act as transmembrane receptors for netrin-1 and play a crucial role in axon guidance and migration of neural cells. Additionally, when cleaved by a caspase to produce an intracellular fragment containing a death domain, UNC5H receptors induce apoptosis. This activity is blocked by the binding of netrin-1. In the absence of netrin-1, UNC5H receptors act as tumour suppressors by inhibiting anchorage-independent growth and invasion, but mutation of these receptors provides a potential mechanism for tumourigenicity. The expression of UNC5H receptors is down-regulated in multiple carcinomas, including colorectal, breast, ovary, uterus, stomach, lung, and kidney cancers. UNC5H4, also known as UNC5D (unc-5 homolog D), is single-pass type I membrane protein that is a member of the UNC5H netrin receptor family. Two isoforms of UNC5H4 exist due to alternative splicing events.
