PTH/Parathyroid hormone is a hormone produced by the parathyroid gland that regulates the concentration of calcium and phosphorus in extracellular fluid. This hormone elevates blood Ca2+ levels by dissolving the salts in bone and preventing their renal excretion. It is produced in the parathyroid gland as an 84 amino acid single chain polypeptide. It can also be secreted as N-terminal truncated fragments or C-terminal fragments after intracellular degradation, as in case of hypercalcemia. Defects in this gene are a cause of familial isolated hypoparathyroidism (FIH); also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterised by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. FIH exist both as autosomal dominant and recessive forms of hypoparathyroidism.

This antibody recognises a carbohydrate epitope on a single chain, transmembrane, heavily glycosylated protein of 90 to 120 kDa, which is identified as CD34 (VI international workshop on human differentiation antigens). Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, it is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. CD34 expression is also found in vascular endothelium. Additionally, proliferating endothelial cells overexpress this molecule than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi s sarcoma, but shows low specificity.

Recognises a single chain glycoprotein of 105/120 kDa, identified as CD30/Ki-1. CD30 is synthesised as a 90 kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120 kDa glycoprotein. In Hodgkin s disease, CD30/Ki-1 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. It is also expressed by the tumor cells of a majority of anaplastic large cell lymphomas as well as by a varying proportion of activated T and B cells. This mAb distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas. About one third of the Ki-1 positive lymphomas lack the leukocyte common antigen (CD45).

Prolactin is a peptide hormone of pituitary origin and is well known for the stimulation, initiation, and maintenance of lactation. Prolactin is also known to regulate mammary gland development and immune system, and has an essential role in metabolism and behavioral modification (Bernichtein <i>et al.</i>). Prolactin actions are mediated by the prolactin receptor (PRLR), which is a single-chain transmembrane protein composed of an extracellular, transmembrane, and intracellular domain. It has also been indicated that high to normal circulating levels of prolactin increase breast cancer risk, and dopamine agonists have been suggested to be effective for treatment (Bernichtein <i>et al.</i>). Prolactin levels have been reported to be higher in multiple sclerosis patients, which promotes B cell autoreactivity (Correale <i>et al.</i>). It has been shown that administration of recombinant prolactin after bone marrow transplantation in mice promotes immune and myeloid reconstitution (Sun <i>et al.</i>).

Three isoforms of NCAM/CD56 are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. NCAM antibody recognises two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). NCAM antibody plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.
