Recognizes human ORAI3, also known as Transmembrane protein 142C (TMEM142C), a 31.5 kDa multi-pass membrane protein belonging to the ORAI family. Antigen stimulation of immune cells triggers Ca2+ entry through Ca2+ release-activated Ca2+ (CRAC) channels. CRAC channels are the main pathway for Ca2+ influx in T-cells and promote the immune response to pathogens by activating the transcription factor NFAT. ORAI3 is one of two mammalian homologs of ORAI1, a four- transmembrane spanning protein that is an essential component of CRAC channels. ORAI3, along with ORAI1 and ORAI2, functions as a Ca2+ plasma membrane channel that is gated through interactions with STIM1, the store-activated endoplasmic reticulum Ca2+ sensor. Studies indicate that ORAI3 channels undergo a lesser degree of depotentiation than ORAI1 or ORAI2. Is not expected to cross react with either ORAI1 or ORAI2.

STIM1 Antibody: In T lymphocytes, the sole pathway for Ca++ entry following antigen-receptor binding is through store-operated Ca++-release-activated Ca++ (CRAC) channels. These channels are made up of the pore-forming subunit ORAI1 and the stromal interaction molecule 1 (STIM1), a protein that functions as a Ca++ sensor and activates the CRAC channels, migrating to the plasma membrane from endoplasmic reticulum (ER)-like sites which act as the Ca++ store. A related molecule, STIM2, acts to inhibit the STIM1-mediated store-operated Ca++ entry, and can form complexes with STIM1, suggesting they may play a coordinated role in controlling Ca++ entry. At least two isoforms of STIM1 are known to exist; this antibody will detect only the larger form. This STIM1 antibody is predicted to have no cross-reactivity to STIM2.

ATP2C2 Antibody: ATP2C2, also known as secretory pathway Ca2+/Mn2+-ATPase (SPCA) 2, belongs to the family of P-type cation transport ATPases. This magnesium-dependent enzyme catalyzes the hydrolysis of ATP coupled with the transport of the calcium from the cytosol to the Golgi lumen. Defects in the related gene ATP2C1 cause Hailey-Hailey disease, for which ATP2C2 does not compensate, suggesting that ATP2C2 plays other physiological roles. Unlike ATP2C1, ATP2C2 has a much more restricted expression pattern and displays a higher maximal turnover rate for overall Ca2+-ATPase reaction and a lower apparent affinity for cytosolic Ca2+ activation of phosphorylation. Overexpression of ATP2C2 in mammary tumors result a Ca2+ influx via the store-operated Ca2+ channel ORAI1 and independent of the STIM1 and STIM2 sensors.

TLR11, an innate sensor for profilin (Toxoplasma gondii), is an intracellular receptor that resides in the endoplasmic reticulum. The 12 membrane-spanning endoplasmic reticulum-resident protein UNC93B1 interacts directly with TLR11 and regulates the activation of dendritic cells in response to profilin (Toxoplasma gondii) and parasitic infection in vivo. TLR12, a previously uncharacterised TLR, also recognises profilin from Toxoplasma gondii. TLR12 is sufficient for recognition of profilin by plasmacytoid dendritic cells (pDCs), whereas TLR11 and TLR12 are both required in macrophages and conventional DCs. In contrast to TLR11, TLR12-deficient mice succumb rapidly to T. gondii infection.

DDX60 Antibody: DDX60 is a DEXD/H Box RNA helicase and the human homolog of the S. cerevisiae Ski2, a cofactor of RNA exosome. Expression of DDX60 increases after viral infection and binds to endogenous RIG-I, a cytoplasmic viral RNA sensor, as well as other RIG-I-like receptors such as MDA5 and LGP2. These proteins activate TBK1 and IKK-e, signaling IRF3 and NF-kB-responsive genes in the innate immune response pathway. DDX60 is required for the RIG-I- or MDA5-dependent response to viral infection, but is dispensable for TLR3-mediated signaling.
