Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH17 is a 1,159 amino acid single-pass type I membrane protein that contains six cadherin domains. Expressed as multiple alternatively spliced isoforms, PCDH17 is thought to function as a calcium-dependent cell adhesion protein that may play a role in establishing cell-cell connections within brain tissue. The gene encoding PCDH17 maps to human chromosome 13, which houses over 400 genes, such as BRCA2 and RB1, and comprises nearly 4% of the human genome.

Angelman syndrome, Prader-Willi syndrome, Tay-Sachs disease and Marfan syndrome are all associated with defects in chromosome 15-localized genes.

LRFN2 is a 789 amino acid single-pass type I membrane protein belonging to the LRFN family. Encoded by a gene that maps to human chromosome 6p21.2, LRFN2 is moderately expressed in brain, spleen and testis. LRFN2 contains one fibronectin type-III domain, one Ig-like (immunoglobulin-like) domain and six LRR (leucine-rich) repeats. LRFN2 promotes neurite outgrowth in hippocampal neurons, enhances cell surface expression of two NMDA receptor subunits, NMDAΩ1 and NMDAé1, and may play a role in redistributing PSD-95 to cell periphery. LRFN2 forms heteromeric complexes with LRFN1, LRFN3, LRFN4 and LRFN5, and is capable of forming homomeric complexes, but not across cell junctions.

Thyroxine Binding Globulin (TBG) is one of three proteins responsible for carrying the thyroid hormones thyroxine (T4) and 3,5,3 triiodothyronine (T3) in the bloodstream, along with transthyretin and albumin. TBG has the highest affinity for T4 and T3, but is present in the lowest concentration of the three proteins. Despite its low concentration, TBG carries the majority of T4 in serum. Due to the very low serum concentration of T4 & T3, TBG is rarely more than 25% saturated with ligand. Unlike transthyretin and albumin, TBG has a single binding site for T4/T3. Genomically, TBG is a serpin, although it has no inhibitory function like many other members of this class of proteins. TBG is synthesized primarily in the liver.

Staphylococcal enterotoxins represent a group of proteins, which are secreted by Staphylococcus aureus and cause staphylococcal food poisoning syndrome. The illness is characterised by high fever, hypotension, diarrhea, shock, and in some cases death. Their molecular masses range between 27 and 30 kDa. At present, seven enterotoxins are known, namely A, B, C1, C2, C3, D and E. Their amino acid sequences have been determined and it was shown that all are single chain polypeptides containing one disulfide bond formed by two half cystines located in the middle of the polypeptide chain, which form the so called cysteine loop. SEB is an extremely potent activator of T cells, stimulating the production and secretion of various cytokines which mediate many of the toxic effects of SEB.

The amphoterin-induced gene and ORF (AMIGO) family of proteins consists of AMIGO1, AMIGO2 and AMIGO3. All three members are single pass type I membrane proteins that contain several leucine-rich repeats, one IgG domain, and a transmembrane domain. The AMIGO proteins are specifically expressed on fiber tracts of neuronal tissues and participate in their formation. The AMIGO proteins can form complexes with each other, but can also bind itself. AMIGO1, also designated Alivin-2, promotes growth and fasciculation of neurites and plays a role in myelination and fasciculation of developing neural axons. In cerebellar neurons, AMIGO2 (Alivin-1) is crucial for depolarization-dependent survival. Similar to AMIGO1 and AMIGO2, AMIGO3 (Alivin-3) plays a role in homophilic and/or heterophilic cell-cell interaction and signal transduction.

Many growth factors function by binding receptors with intrinsic tyrosine kinase activity (1,2). Signaling by such receptors involves a series of intermediates characterized by SH2 domains that bind tyrosine phosphorylated receptors by a direct interaction between the SH2 domain and the phosphotyrosine-containing receptor sequences (3,4). GRB7, a SH2 domain protein, has a single SH2 domain at its C-terminal, a central region with similarity to Ras GAP and a proline-rich N-terminus (5,6). GRB7 maps to the region on mouse chromosome 11 containing the Neu gene (6). This region of mouse chromosome 11 is syntenic to an area of human chromosome 17q that is frequently amplified in breast cancer (6,7). Moreover, GRB7 is amplified and over-expressed in breast cancer and is found in a complex with Neu gp185 (6).

Neuronal ceroid-lipofuscinose (NCL), also designated Batten disease, comprises a group of recessively inherited, progressive neurodegenerative diseases found in children. NCL is characterized by atrophy of the brain and an accumulation of lysosome derived fluorescent bodies found in many cells, especially neurons. Symptoms of NCL include a failure of psychomotor development, seizures, impaired vision and premature death. The eight genes/proteins associated with NCL are designated CLN1-CLN8. Mutations in six of these genes results in a distinct type of NCL-disease; the six genes/proteins are CLN1 (encoding PPT1, a protein thiolesterase), CLN2 (encodeing the serine protease TPP1), CLN3, CLN5, CLN6 and CLN8. A single base duplication mutation in dog and cow CLN5 has been shown to cause NCL.

EphB1, previously known as Elk (eph like kinase), is a receptor tyrosine kinase of the highly tissue restricted family of eph proteins. EphB1 and other ephB family members are type 1 membrane spanning proteins, comprised of immunoglobulin, fibronectin type III, and cysteine rich subdomains in the ecto domain, and the single uninterrupted cytoplasmic tyrosine kinase domain upstream of a carboxyterminal sterile alpha motif (SAM) domain. EphB family proteins bind ephrins of the B class. EphB1 is expressed predominately in developing neural structures in embryos, and in vascular epithelium of kidney, and other tissues. Upon binding to alternatively oligomerized ephrin B1, EphB1 signals regulation of cell attachment and cell to cell assembly. Members of this protein family are implicated in neuronal and vascular cell targeting.
