APC8 Antibody: Cell cycle regulated protein ubiquitination and degradation within subcellular domains is thought to be essential for the normal progression of mitosis. APC8 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. In Drosophila, silencing of APC8 results in developmental delay and pupal lethality with elevated levels of apoptosis, high mitotic index, and delayed or blocked mitosis.

IGF-BPs form high affinity complexes with both IGF-I and IGF-II and act to control of the distribution, function and activity of IGFs in various cell tissues and body fluids. There are seven named IGF-BPs. IGF-BP7 plays a role in skeletal myogenesis by binding to IGF in a manner that inhibits IGF induced differentiation of skeletal myoblasts, without affecting IGF induced proliferation. Additionally, IGF-BP7 suppresses growth and colony formation of prostate and breast cancer cell lines through an IGF independent mechanism, which causes a delay in the G1 phase of the cell cycle, and increased apoptosis. IGF-BP7 is expressed in a wide range of normal human tissues and it usually shows reduced expression in cancer cell lines of prostate, breast, colon, and lung origin.

Voltage-gated potassium (Kv) channels represent the most complex class of voltage-gated ion channels from both functional and structural standpoints. Their diverse functions include regulating neurotransmitter release, heart rate, insulin secretion, neuronal excitability, epithelial electrolyte transport, smooth muscle contraction, and cell volume. Four sequence-related potassium channel genes - shaker, shaw, shab, and shal - have been identified in Drosophila, and each has been shown to have human homolog(s). This gene encodes a member of the potassium channel, voltage-gated, shal-related subfamily, members of which form voltage-activated A-type potassium ion channels and are prominent in the repolarization phase of the action potential. This gene is expressed at moderate levels in all tissues analyzed, with lower levels in skeletal muscle.

PNAS4 Antibody: PNAS4 is a highly conserved protein that shares high homology from plants to animals. It was initially identified as a putative apoptosis-related protein in the human acute promyelocytic leukemia cell line NB4. PNAS4 is activated during the early response to DNA damage and can inhibit proliferation via apoptosis when overexpressed in some tumor cells such as U2OS, SKOV3, and A549. PNAS4 inhibits tumor cell proliferation through the following mechanisms: (i) overexpression of PNAS4 causes S phase arrest by regulating the expression of cell cycle-related proteins and (ii) PNAS4 induces apoptosis through the mitochondrial apoptosis pathway. Recent evidence has shown that PNAS4 may be involved in the genesis of some cancers and could be a potential candidate for lung cancer therapy alone or in combination with gemcitabine. At least two isoforms of PNAS4 are known to exist.

MultimericTNF-alpha™ is a high activity construct in which two trimeric TNF-alpha ligands are artificially linked via the collagen domain of ACRP30. The receptor TNF-R1 is activated by both the membrane-bound and soluble trimeric forms of TNF-alpha, whereas the receptor TNF-R2 only responds to the membrane-bound form of TNF-alpha (TNF-alpha (human) (multimeric) (rec. ) (Prod. No. AG-40B-0019). Since the MultimericTNF-alpha™ mimics the membrane-bound form (forms oligomers higher than trimer), it is the only TNF-alpha protein that can activate the TNF-R2. For TNF-R1 activation, either "normal" TNF-alpha or MultimericTNF-alpha™ can be used.

Substrate-specific adapter of a DCX (DDB1-CUL4-X-box) E3 ubiquitin-protein ligase complex required for cell cycle control, DNA damage response and translesion DNA synthesis. The DCX(DTL) complex, also named CRL4(CDT2) complex, mediates the polyubiquitination and subsequent degradation of CDT1 and CDKN1A/p21(CIP1). CDT1 degradation in response to DNA damage is necessary to ensure proper cell cycle regulation of DNA replication. CDKN1A/p21(CIP1) degradation during S phase or following UV irradiation is essential to control replication licensing. Most substrates require their interaction with PCNA for their polyubiquitination: substrates interact with PCNA via their PIP-box, and those containing the 'K+4' motif in the PIP box, recruit the DCX(DTL) complex, leading to their degradation. In undamaged proliferating cells, the DCX(DTL) complex also promotes the 'Lys-164' monoubiquitination of PCNA, thereby being involved in PCNA-dependent translesion DNA synthesis.

Description:
Interleukin-22 (IL-22) was initially identified as a gene induced by IL-9 in mouse T cells and mast cells. Mouse IL-22 cDNA encodes a 179 amino acid residue protein with a putative 33 amino acid signal peptide that is cleaved to generate a 147 amino acid mature protein that shares approximately 79% and 22% sequence identity with human IL22 and IL10, respectively. IL22 has been shown to activate STAT-1 and STAT-3 in several hepatoma cell lines and up-regulate the production of acute phase proteins. IL-22 is produced by normal mouse T cells upon Con A activation. Mouse IL-22 expression is also induced in various organs upon lipopolysaccharide injection, suggesting that IL-22 may be involved in inflammatory responses. The functional IL-22 receptor complex consists of two receptor subunits, IL-22R (previously an orphan receptor named CRF2-9) and IL-10R beta (previously known as CRF2-4), belonging to the class II cytokine receptor family.
