SCRG1 (scrapie-responsive protein 1) is a 98 amino acid secreted protein that belongs to the SCRG1 family. The SCRG1 protein contains a 20-amino acid signal peptide, and is expressed abundantly in the central nervous system of human adult, but not at all in fetal brain. The protein is targeted to the Golgi apparatus and large dense-core vesicles/secretory granules in neurons. High levels of SCRG1 transcripts are also observed in testis and aorta. SCRG1 is associated with neurodegenerative changes observed in transmissible spongiform encephalopathies. It may play a role in host response to prion-associated infections. The SCRG1 protein may be partly included in the membrane or secreted by the cells due to its hydrophobic N-terminus. The human and mouse SCRG1 proteins share 83% sequence identity. The SCRG1 gene is conserved in chimpanzee, cow, mouse, rat and chicken, and maps to human chromosome 4q34.1.

The apolipoprotein L gene family encodes six highly homologous proteins designated apoL-I to -VI, which are associated with large high density type lipoproteins (HDL) (2,3). The human apoL family maps to chromosome 22q12.1-13.1 within a 127,000-bp region (4). ApoL has been characterized as a pancreas specific, 383-amino acid protein that contains a 12-amino acid secretory signal peptide (4). The apoL genes have TATA-less promoters and contain putative sterol regulatory elements, suggesting that transcription of these genes may be coordinated with that of the low density lipoprotein receptor and genes in pathways involving the synthesis of triglycerides and cholesterol (3). ApoL homologs can undergo 10 fold changes in expression during atherosclerotic changes in vascular endothelial cells, which includes the inflammatory reaction of atherosclerotic lesions (5).

Protocadherins are a subfamily of cadherins, a large group of related glycoproteins that mediate calcium-dependent cell-to-cell adhesion via a homophilic mechanism. Involved in a variety of functions, protocadherins help to regulate neural development and synapse formation. PCDH1 is a 1,026 amino acid single-pass type I membrane protein that contains seven cadherin domains and is a member of the protocadherin family. Localized to cell-cell and cell-matrix boundaries and expressed at high levels in brain and neuro-glial cells, PCDH1 is thought to be involved in cell adhesion and cell-cell interactions and may play a role in neuronal development. PCDH1 contains a C-terminal cytoplasmic region, an extracellular region and a transmembrane region, and is expressed as two isoforms due to alternative splicing events.

Staphylococcal enterotoxins represent a group of proteins, which are secreted by Staphylococcus aureus and cause staphylococcal food poisoning syndrome. The illness is characterised by high fever, hypotension, diarrhea, shock, and in some cases death. Their molecular masses range between 27 and 30 kDa. At present, seven enterotoxins are known, namely A, B, C1, C2, C3, D and E. Their amino acid sequences have been determined and it was shown that all are single chain polypeptides containing one disulfide bond formed by two half cystines located in the middle of the polypeptide chain, which form the so called cysteine loop. SEB is an extremely potent activator of T cells, stimulating the production and secretion of various cytokines which mediate many of the toxic effects of SEB.

ZNRF2 (zinc and ring finger 2), also known as RNF202, is a 242 amino acid peripheral membrane protein that contains one RING-type zinc finger and localizes to the lysosome, as well as the endosome and the cell junction. Expressed at high levels in brain tissue, ZNRF2 is thought to function as an E3 ubiquitin-protein ligase that may be involved in the establishment and maintenance of neuronal transmission and plasticity. Upon DNA damage, ZNRF2 is subject to phosphorylation, probably by ATR or ATM.

This antibody cocktail recognizes acidic (Type I or LMW) and basic (Type II or HMW) cytokeratins, which include CK1, CK3, CK4, CK5, CK6, CK8, CK10, CK14, CK15, CK16, and CK19. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. KRTL/KRTH is a broad spectrum anti pan-cytokeratin antibody cocktail, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer. It has been used to characterize the source of various neoplasms and to study the distribution of cytokeratin containing cells in epithelia during normal development and during the development of epithelial neoplasms. This antibody stains cytokeratins present in normal and abnormal human tissues and has shown high sensitivity in the recognition of epithelial cells and carcinomas.

TSKS is a 592 amino acid protein that is highly expressed in human testicular tissue. Low levels of TSKS are detectable in prostate, placenta, fetal liver, thymus, and mammary gland tissues. TSKS is found to be downregulated in cancerous testicular tissue from seminoma, teratocarcinoma, embryonal and Leydig cell tumors concurrently with high expression in neighboring premalignant carcinoma. TSKS protein contains an N-terminal signal peptide, but does not contain a transmembrane region. TSKS has many potential phosphorylation and glycosylation sites and is phosphorylated by soluble recombinant TSSK2 in vitro. It is thought that TSKS likely plays a physiological role in spermatogenesis or spermiogenesis.

A schizophrenia susceptibility locus has been identified on chromosome 22 and studies show that 22q11 deletion symptoms include a high incidence of schizophrenia. Cdc123 (Cell division cycle protein 123), also known as D123, is a 336 amino acid cytoplasmic protein that is involved in cell cycle control. Widely expressed with high expression in thymus, spleen, ovary, testis, small intestine and leukocytes, Cdc123 functions to destabilize Chfr (checkpoint with forkhead and ring finger domain) proteins which, when accumulated, block the G to S phase transition. Cdc123 prevents the Chfr proteins from collecting in the cell, thereby allowing the cell to enter the S phase. Due to its role in cell cycle control, Cdc123 is thought to be a basal marker for luminal breast cancers.

UCH-L1 is a member of a gene family whose products hydrolyze small C-terminal adducts of ubiquitin to generate the ubiquitin monomer. Expression of UCH-L1 is highly specific to neurons and to cells of the diffuse neuroendocrine system and their tumors. UCH-L1 is expressed in brain neurons. Examination of specific brain regions reveals expression in all areas tested, particularly in the substantia nigra. UCH-L1 represents 1 to 2% of total soluble brain protein. Its occurrence in Lewy bodies and its function in the proteasome pathway make it a compelling candidate gene in Parkinson disease. The gene which encodes UCH-L1 maps to human chromosome 4p14. The 230 amino acid human UCH-L3 protein is 54% identical to that of UCH-L1. UCH-L3 is the predominant thiol protease and has high-affinity binding sites for ubiquitin.

ACO2, also referred to as aconitate hydratase, citrate hydrolyase or aconitase, is an iron-sulfur hydrolyase that catalyzes the non-limiting interconversion of citrate and isocitrate in the tricarboxylic acid cycle. It is expressed in the mitochondria and maintains a citrate:isocitrate ratio of approximately 10:1. ACO2 contains a redox-sensitive iron-sulfur cluster that exists in two states: active (Fe4S4) and inactive (Fe3S4). ACO2 activity is dependent on the state of this cluster as well as the presence of two conserved cysteine residues. In normal prostate epithelial cells ACO2 activity is prevented due to the high levels of zinc inhibiting the enzyme. In these citrate-producing epithelial cells citrate oxidation is impaired allowing citrate to accumulate and exhibit a citrate:isocitrate ratio of approximately 30:1. In malignant prostate cells zinc is unable to accumulate, therefore ACO2 activity resumes and citrate is oxidized.

Belonging to the glycosyltransferase 14 family, GCNT7, also known as Beta-1,3-galactosyl-O-glycosyl-glycoprotein beta-1,6-N-acetylglucosaminyltransferase 7, is a 430 amino acid glycosyltransferase that is localized to the Golgi apparatus. Other members of this family include GCNT1, GCNT2, GCNT3, GCNT4 and GCNT6. GCNT1 has been shown to play an important regulatory role in the biosynthesis of mucin-type O-glycans, which serve as ligands in cell adhesion. Specifically, GCNT1 expression in leukocytes regulates the synthesis of core 2 O-glycans that carry sialyl-Lewis x (sLex) oligosaccharides, which confer high affinity binding to Selectin proteins. Since downregulation of Selectin ligand expression has been shown to inhibit tissue infiltration, glycosyltransferase 14 family members represent potential drug targets for the treatment of inflammatory disorders and other pathologies involving Selectin proteins.
