Members of the transforming growth factor-beta (TGF-Beta) superfamily play critical roles in controlling cell growth and differentiation. Effects of TGF-Beta family ligands are mediated by Smad proteins. The Smad nuclear interacting protein (SNIP1) contains a forkhead-associated (FHA) domain and acts as a nuclear inhibitor of CBP/p300. SNIP1 potently inhibits the activity of NF-kappa B, which binds the C/H1 domain of CBP/p300, by competing for the binding site. SNIP1 is also thought to induce expression of Cyclin D1 to promote cellular proliferation. SNIP1 is ubiquitously expressed with high expression in heart and skeletal muscle.

C5L2 is an integral membrane protein that belongs to the Gprotein-coupled receptor 1 family. C5L2 weakly couples to Gi-mediated signaling pathways and is widely expressed in the hippocampus, hypothalamus, pons, frontal cortex and liver. It is coexpressed with the C5a receptor (C5AR) on polymorphonuclear neutrophils and may modulate the activity of C5AR. The function of the C5a receptors, particularly that of C5L2, is still unclear. This gene encodes a protein that is expressed at high levels in cardiac and skeletal muscle. Mutations in this gene have been associated with a number of clinical disorders including Barth syndrome, dilated cardiomyopathy (DCM), hypertrophic DCM, endocardial fibroelastosis, and left ventricular noncompaction (LVNC). Multiple transcript variants encoding different isoforms have been described. A long form and a short form of each of these isoforms is produced; the short form lacks a hydrophobic leader sequence and may exist as a cytoplasmic protein rather than being membrane-bound. Other alternatively spliced transcripts have been described but the full-length nature of all these transcripts is not known.

Collagen proteins (products of the COL gene family) are fibrous, extracellular matrix proteins with high tensile strength and are the major components of connective tissue such as tendons and cartilage. All collagens contain a triple helix domain and frequently show lateral self-association in order to form complex connective tissues. The Collagen Type XXV precursor produces the proteolytic fragment CLAC (collagenous Alzheimer amyloid plaque component), which deposits in senile plaques associated with amyloid beta peptides (Abeta) in the brains of patients with Alzheimer's disease. CLAC binds to the fibrillized form of Abeta, which disturbs the structure and function of plasma membranes.

For example, the ability to identify sweet-tasting foods enables animals to seek out food with high nutritive value, whereas the ability to identify bitter substances enables them to avoid the ingestion of potentially harmful substances. A family of integral membrane proteins are involved in taste perception and include T1R, which is involved in sweet taste perception and T2R, which is involved in bitter taste perception. Both types of taste receptors couple to various G proteins to initiate signal transduction cascades. Specifically, T2R50 is expressed in subsets of taste receptor cells of the tongue and exclusively in gustducin-positive cells.

BAMBI is a membrane spanning glycoprotein that acts as a negative regulator of TGF-beta signaling during development. The BAMBI family of proteins are related to type I TGF-beta receptor family, however, BAMBI is a pseudoreceptor that lacks an intracellular serine/threonine kinase domain. BAMBI transcription regulation is under the influence of beta-catenin, BMP, smad3 and smad4. BAMBI expression can increase in colorectal and hepatocellular carcinomas relative to non-cancerous tissues. BAMBI is expressed at high levels during odontogenesis. BAMBI is coexpressed with Bmp-4 during early Xenopus embryogenesis and can be detected in poorly metastatic human melanoma cell lines.
