Kaposi's sarcoma-associated herpesvirus (KSHV) belongs to the gamma-(2)-herpesvirus subfamily and has been closely linked to the Kaposi’s sarcoma, primary effusion lymphoma (PEL) and multicentric Castleman’s disease. The genome of KSHV is 165-170 kb and contains at least 88 open reading frames. ORF45 protein encoded by an immediate-early gene in the KSHV genome is characterized as a phosphorylated protein, and it is localized in the cytoplasm of infected cells. Studies have shown that ORF45 Protein interacted with cellular IRF-7 and blocked virus-mediated phosphorylation and nuclear translocation of IRF-7. In consequence, ORF45 efficiently inhibited virus-induced production of type I IFN. Zhu et al (2003) reported that the ORF45 Protein was associated with purified virions.

Mouse ULBP1, also known as RAET1I and NKG2DL1, is a member of the ULBP/RAET1 gene family. ULBP1 plays an important role in immune responses, especially in cancer and infectious diseases, and is well-known to bind to NKG2D together with at least ULBP 2 and 3. These proteins are distantly related to major histocompatibility class I (MHC I) molecules, possessing the alpha 1 and alpha 2 Ig-like domains, but lacking the alpha 3 domain. Unlike MHC Class I, they have no capacity to bind peptide or interact with beta2-microglobulin. It can activate multiple signalling pathways in primary NK cells, gamma delta T cells, and CD8+ alpha beta T cells, resulting in the production of cytokines and chemokines.ULBP1 is expressed in wide range of tissues including heart, brain, lung, liver, bone marrow and some tumour cells, T-cells, B-cells, As an unconventional member of the MHC class I family, ULBP1 is able to interact with soluble CMV glycoprotein UL16 in CMV infected cells. The interaction with UL16 blocked the interaction with the NKG2D receptor, and thus might escape the immune surveillance. Furthermore, UL16 also causes ULBP1 to be retained in the ER and cis-Golgi apparatus so that it does not reach the cell surface. The ULBP1 regulation may have implications for development of new therapeutic strategies against cancer cells.

Gentamicin is an aminoglycoside antibiotic complex produced by fermentation of Micromonospora purpurea or M. echinospora. It is a mixture of 3 major components designated as C1, C1a, and C2. Gentamicin is used as the sulfate salt. Each component consists of five basic nitrogens and requires five equivalents of sulfuric acid per mole of gentamicin base.
Mode of action: Gentamicin causes codon misreading by binding to the 30S ribosomal subunit, blocking the translocation of peptidyl-tRNA from the acceptor site to the donor site.The bactericidal effect of gentamicin on Pseudomonas aeruginosa is exerted by the binding of gentamicin to the outer membrane, where it displaces natural cations, destabilizes the membrane, and forms holes in the cell surface.Antimicrobial spectrum: Gram-negative bacteria, Staphylococcus aureus and other Gram-positive bacteria. Gentamicin sulfate is a broad spectrum antibiotic. It inhibits the growth of a wide variety of Gram-positive and Gram-negative microorganisms, including strains resistant to tetracycline, chloramphenicol, kanamycin, and colistin, particularly strains of Pseudomonas, Proteus, Staphylococcus, and Streptococcus. Gentamicin sulfate inhibits bacterial protein biosynthesis by binding to the 30S subunit of the ribosome.

The J43.1 monoclonal antibody specifically reacts with mouse CD279, also known as PD-1 (programmed death-1), a 50 to 55 kDa glycoprotein of the Ig superfamily. The PD-1 ligands, PD-L1 (B7-H1) and PD-L2 (B7-H2) are members of the B7 family. Pd-1 contains an Immunoreceptor Tyrosine-based Inhibitory Motif (ITIM) and influences the peripheral tolerances and autoimmune diseases in mice. PD-1 is transiently expressed on CD4/CD8 thymocytes, it is upregulated in apoptotic cells, and it is expressed by activated myeloid and T and B cells. The binding of PD-1 to its ligands is blocked by the J43 antibody, which also enhances contact hypersensitivity and exacerbates acute Graft-versus-host disease, Experimental autoimmune encephalomyelitis and NOD diabetes. PD-1 seems to downregulate the immune response, as the development of splenomegaly and breakdown of peripheral tolerance in PD-1 deficient mice suggests.
