Description:   p53, a DNA-binding, oligomerization domain- and transcription activation domain-containing tumor suppressor, upregulates growth arrest and apoptosis-related genes in response to stress signals, thereby influencing programmed cell death, cell differentiation, and cell cycle control mechanisms. p53 localizes to the nucleus, yet can be chaperoned to the cytoplasm by the negative regulator, MDM2. MDM2 is an E3 ubiquitin ligase that is upregulated in the presence of active p53, where it poly-ubiquitinates p53 for proteasome targeting. p53 fluctuates between latent and active DNA-binding conformations and is differentially activated through posttranslational modifications, including phosphorylation and acetylation. Mutations in the DNA-binding domain (DBD) of p53, amino acids 110-286, can compromise energetically-favorable association with cis elements and are implicated in several human cancers.

Description:   p44/42 MAP Kinase(Thr202); ERK (extracellular signal regulated kinase), also known as MAPK (mitogen activated protein kinase) has two closely related isoforms of 44 kDa and 42 kDa, respectively. These kinases belong to a family of serine/threonine kinases that are activated upon treatment of cells with a large variety of stimuli including mitogens, hormones, growth factors, cytokines, and bioactive peptides. Cell stimulation induces the activation of a signaling cascade, the downstream effects of which have been linked to the regulation of cell growth and differentiation as well as the cytoskeleton. ERK1 and ERK2 are phosphorylated within the activation loop on both a Threonine and a Tyrosine residue (within a Thr-Glu-Tyr motif) by MEKs (MAPK/ERK kinases), thereby greatly elevating the activity of ERK1&2.

Description:   Angiotensin Converting enzyme is involved in catalyzing the conversion of angiotensin I into a physiologically active peptide angiotensin II. Angiotensin II is a potent vasopressor and aldosterone-stimulating peptide that controls blood pressure and fluid-electrolyte balance. This enzyme plays a key role in the renin-angiotensin system. ACE converts angiotensin I to angiotensin II by release of the terminal His-Leu, this results in an increase of the vasoconstrictor activity of angiotensin. Also able to inactivate bradykinin, a potent vasodilatator. ACE exists in two forms, a 170KD somatic form and a 90KD germinal form. The somatic form is expressed by endothelial cells (especially those of lung capillaries and arterioles), epithelial cells (especially in proximal renal tubules and small intestine), by some neuronal cells and variably by some macrophages and T lymphocytes. The germinal form is expressed by spermatozoa.

Description:   Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH8 (protocadherin-8), also known as Arcadlin or PAPC, is a 1,070 amino acid single-pass type I membrane protein that contains six cadherin domains and belongs to the protocadherin family. Localized to the cell membrane and expressed specifically in fetal and adult brain, PCDH8 is thought to play a role in cell adhesion events in the central nervous system (CNS). PCDH8 is inactivated or silenced in breast cancer, suggesting a possible role in tumor suppression. Two isoforms of PCDH8 that differ in their cytoplasmic tails are expressed due to alternative splicing events.

Description:   Epithelial membrane protein-1 (EMP-1) is a four pass transmembrane protein consisting of 160 amino acids. It is a member of a small family of epithelial membrane proteins. EMP-1 is very similar in structure to its close relative, Peripheral Myelin Protein 22 (PMP22). It is most predominantly expressed in tissues of the gastrointestinal tract but has also been found to be a junctional protein in the liver expressed along the intercellular border. EMP-1 directly interacts with the C-terminus of the P2X7 receptor and may be involved in membrane blebbing. EMP-1 may also be an important regulator in cell communication, signaling, and adhesion. When EMP-1 is overexpressed, cell proliferation is inhibited, S phase is arrested and G1 phase is prolonged in esophogeal cancer cells. EMP-1 may play a role in tumorigenesis and has been identified as a biomarker for gefitinib treatment resistance for patients with lung cancer.

Description:   Thrombopoietin (TPO or THPO), also known as c-Mpl ligand (c-Mpl L), is a cytokine that plays a central role in megakaryopoiesis by influencing the development and maturation of megakaryocytes and platelet production from hematopoietic stem cells. TPO exerts its biological effects through the TPO receptor, c-Mpl. c-Mpl is a member of the cytokine receptor superfamily. Expression of c-Mpl is restricted to hematopoietic tissues and cells, such as bone marrow, spleen, fetal liver and CD34+ cells. Stimulation of c-Mpl with TPO results in the activation of the Janus tyrosine kinase family members, Tyk 2 and JAK2, which in turn phosphorylate Stat5 and Stat3, causing their nuclear translocation and the transcription of Stat responsive genes. Muta-tions in c-Mpl have been implicated as the cause of certain human disorders, including congenital amegakaryocytic thrombocytopenia (CAMT) and thrombocytopenia with absent radii (TAR) syndrome.

Description:   TWA1 is a 228 amino acid nuclear protein that is highly conserved throughout evolution. TWA1 interacts with Ran BP-M, a protein that is implicated in a diverse array of cellular processes including DNA replication, entry into and exit from mitosis, and the transport of RNA and proteins through the nuclear pore complex.
