Glycerol is used both in sample preparation and gel formation for polyacrylamide gel electrophoresis. Glycerol (5-10%) increases the density of a sample so that the sample will layer at the bottom of a gel’s sample well. Glycerol is also used to aid in casting gradient gels and as a protein stabilizer and storage buffer component. Glycerol is used in the concentration and storage of enzymes. Also prevents back-diffusion and protein samples into the buffer. Ideal for use as a cryoprotectant.

Description:
Granulocyte-macrophage colony-stimulating factor (GM-CSF) promotes the proliferation and differentiation of hematopoietic progenitor cells and the generation of neutrophils, eosinophils, and macrophages. In synergy with other cytokines such as stem cell factor, IL-3, erythropoietin, and thrombopoietin, it also stimulates erythroid and megakaryocyte progenitor cells (Barreda <i>et al.</i>). GM-CSF was first purified from the culture of mouse lung tissue after lipopolysaccharide treatment. GM-CSF is produced by multiple cell types, including stromal cells, Paneth cells, macrophages, dendritic cells (DCs), endothelial cells, smooth muscle cells, fibroblasts, chondrocytes, and Th1 and Th17 cells (Francisco-Cruz <i>et al.</i>). The receptor for GM-CSF (GM-CSFR) is composed of two subunits: the cytokine-specific α subunit (GMRα; CD116) and the common subunit βc (CD131) shared with IL-3 and IL-5 receptors (Broughton <i>et al.</i>). GM-CSFR is expressed on hematopoietic cells, including progenitor cells and immune cells, as well as non-hematopoietic cells. GM-CSF is able to stimulate the development of DCs that ingest, process, and present antigens to the immune system (Francisco-Cruz <i>et al.</i>).

Description:
Neurofilaments are the 10nm or intermediate filament proteins found specifically in neurons, and are composed predominantly of three major proteins called NF-L, NF-M and NF-H. NF-M is the neurofilament middle or medium molecular weight polypeptide and runs on SDS-PAGE gels at 145-160 kDa, with some variability across species boundaries. Antibodies to NF-M are useful for identifying neuronal cells and their processes in tissue sections and in tissue culture. NF-M antibodies can also be useful to visualize neurofilament accumulations seen in many neurological diseases, such as Amyotrophic Lateral Sclerosis (Lou Gehrig's disease) and Alzheimer's disease.

Betacellulin is a member of the epidermal growth factor (EGF) family, and signals through EGF receptor and ERBB4. It activates ERK and AKT pathways, which induces neural stem cell proliferation and prevents spontaneous differentiation in culture. Betacellulin stimulates the expansion of neural stem cells, transit-amplifying cells, and neuroblasts derived from subventricular zone and dentate gyrus (Gómez-Gaviro <i>et al.</i>). It is a potent mitogen for retinal pigment epithelial cells and vascular smooth muscle cells. Betacellulin down-regulates E-cadherin expression in ovarian cancer cell lines via MEK/ERK1/2 and PI3K/AKT signaling pathways, thus increasing cell migration (Zhao <i>et al.</i>). It is a modulator of interferon (IFN) response and enhances anti-viral effects of IFN (Al-Yahya <i>et al.</i>). Betacellulin is expressed in pancreatic α cells, β cells, and duct cells. It induces the proliferation of pancreatic cancer cell lines, inhibits apoptosis, promotes the neogenesis of β cells, and converts non-β cells into insulin-producing cells (Kawaguchi <i>et al.</i>; Miyagawa al.; Saito <i>et al.</i>).

Glycine is a non-essential amino acid. Acts as an inhibitory neurotransmitter, particularly in brain stem and medulla. Shown to block NMDA-stimulated dopamine release in fetal mesencephalic cell cultures. Also reported to increase NMDA-receptor mediated synaptic potential in cortical neurons. It does not have a chiral center, so only amino acid with no asymmetric carbon. Therefore, it does not have a D- or L- isomer designation. This material is synthetic. Thrombopoietin (TPO) is a key regulator of megakaryocytopoiesis and thrombopoiesis <i>in vitro</i> and <i>in vivo</i>. TPO stimulates the proliferation and maturation of megakaryocytes and has an important role in regulating the level of circulating platelets in vivo (Bartley <i>et al.</i>; de Sauvage <i>et al.</i>; Foster <i>et al.</i>; Sohma <i>et al.</i>). TPO also promotes the survival, self-renewal, and expansion of hematopoietic stem cells and primitive multilineage progenitor cells. It is commonly used with other cytokines such as stem cell factor (SCF) and Flt3/Flk-2 ligand to promote expansion of primitive hematopoietic cells in culture (Hitchcock and Kaushansky). The TPO receptor, c-Mpl, is expressed at all stages of megakaryopoiesis, from hematopoietic stem and progenitor cells to mature platelets (Ng <i>et al.</i>).

B18R, also known as B19R in the Copenhagen strain of Vaccinia, is a type I interferon (IFN)-binding protein encoded by the B18R open reading frame in the Western Reserve strain of vaccinia virus. B18R exists in two forms, soluble and membrane-bound form and has a broad species specificity. B18R has high affinity for human IFN-alpha and binds rabbit, bovine, rat, pig, and mouse IFN-alpha and IFN-beta. Secreted B18R binds to uninfected and infected cells. B18R presents at the cell surface and protects cells from the antiviral state. It has shown that binding of soluble recombinant B18R protects cultured cells from IFN and allows vaccinia virus replication.
