Non-receptor protein-tyrosine kinase that regulates reorganisation of the actin cytoskeleton, cell polarisation, cell migration, adhesion, spreading and bone remodeling. Plays a role in the regulation of the humoral immune response, and is required for normal levels of marginal B-cells in the spleen and normal migration of splenic B-cells. Required for normal macrophage polarisation and migration towards sites of inflammation. Regulates cytoskeleton rearrangement and cell spreading in T-cells, and contributes to the regulation of T-cell responses. Promotes osteoclastic bone resorption; this requires both PTK2B/PYK2 and SRC. May inhibit differentiation and activity of osteoprogenitor cells. Functions in signaling downstream of integrin and collagen receptors, immune receptors, G-protein coupled receptors (GPCR), cytokine, chemokine and growth factor receptors, and mediates responses to cellular stress. Forms multisubunit signaling complexes with SRC and SRC family members upon activation; this leads to the phosphorylation of additional tyrosine residues, creating binding sites for scaffold proteins, effectors and substrates. Regulates numerous signaling pathways. Promotes activation of phosphatidylinositol 3-kinase and of the AKT1 signaling cascade. Regulates production of the cellular messenger cGMP. Promotes activation of the MAP kinase signaling cascade, including activation of MAPK1/ERK2, MAPK3/ERK1 and MAPK8/JNK1. Promotes activation of Rho family GTPases, such as RHOA and RAC1. Recruits the ubiquitin ligase MDM2 to P53/TP53 in the nucleus, and thereby regulates P53/TP53 activity, P53/TP53 ubiquitination and proteasomal degradation. Acts as a scaffold, binding to both PDPK1 and SRC, thereby allowing SRC to phosphorylate PDPK1 at 'Tyr-9, 'Tyr-373', and 'Tyr-376'. Promotes phosphorylation of NMDA receptors by SRC family members, and thereby contributes to the regulation of NMDA receptor ion channel activity and intracellular Ca(2+) levels. .

This MAb reacts with a 45 kDa glycopeptide, which is a type II membrane glycoprotein with a transmembrane sequence near the NH2terminus. CD38 is a type II transmembrane glycoprotein that is present on early B- and T-cell lineages and activated B- and T-cells but is absent from most mature resting peripheral lymphocytes. CD38 is also found on thymocytes, pre-B cells, germinal center B-cells, mitogen-activated T-cells, monocytes and Ig-secreting plasma cells. CD38 is expressed on CD34 cells. The CD34 CD38- population of hematopoietic stems cells defines the most pluripotent cells (e.g. blast colony forming cells).

The 10F.9G2 antibody is specific for mouse CD274, more commonly known as PD-L1 or B7-H1, which acts as a ligand for the T cell co-regulatory receptor PD-1 (CD279). This interaction modulates T cell antigen receptor (TCR) signaling and therefore T cell activation. PD-L1 binding to PD-1 expressed on CD4- CD8- thymocytes participates in the processes of clonal selection, elimination of autoreactive lymphocytes, and development of tolerance. PD-L1 may also bind PD-1 following the receptor’s inducible expression on activated, mature T cells, where it has been proposed to limit T cell activation. PD-L1 is one of a group of “B7” ligands whose interactions with the CD28 receptor family, also including CTLA-4 (CD152), provide a balance of co-stimulatory /co-inhibitory signaling important in T cell activation, tolerance, and autoimmunity.

Protocadherin beta 13 is a member of the protocadherin beta gene cluster, one of three related gene clusters tandemly linked on chromosome five. The gene clusters demonstrate an unusual genomic organization similar to that of B-cell and T-cell receptor gene clusters. The beta cluster contains 16 genes and 3 pseudogenes, each encoding 6 extracellular cadherin domains and a cytoplasmic tail that deviates from others in the cadherin superfamily. The extracellular domains interact in a homophilic manner to specify differential cell-cell connections. Unlike the alpha and gamma clusters, the transcripts from these genes are made up of only one large exon, not sharing common 3' exons as expected. These neural cadherin-like cell adhesion proteins are integral plasma membrane proteins. Their specific functions are unknown but they most likely play a critical role in the establishment and function of specific cell-cell neural connections.
