CD2 interacts through its amino-terminal domain with the extracellular domain of CD58 (also designated CD2 ligand) to mediate cell adhesion. CD2/CD58 binding can enhance antigen-specific T cell activation. CD2 is a transmembrane glycoprotein that is expressed on peripheral blood T lymphocytes, NK cells and thymocytes. CD58 is a heavily glycosylated protein with a broad tissue distribution in hematopoietic and other cells, including endothelium. Interaction between CD2 and its counter receptor LFA3 (CD58) on opposing cells optimizes immune system recognition, thereby facilitating communication between helper T lymphocytes and antigen-presenting cells, as well as between cytolytic effectors and target cells.

Cadherins are calcium-dependent cell-cell adhesion molecules, and protocadherins constitute a subfamily of nonclassic cadherins. Protocadherin beta 13 is a member of the protocadherin beta gene cluster, one of three related gene clusters tandemly linked on chromosome five. The gene clusters demonstrate an unusual genomic organization similar to that of B-cell and T-cell receptor gene clusters. The beta cluster contains 16 genes and 3 pseudogenes, each encoding 6 extracellular cadherin domains and a cytoplasmic tail that deviates from others in the cadherin superfamily. The extracellular domains interact in a homophilic manner to specify differential cell-cell connections. Unlike the alpha and gamma clusters, the transcripts from these genes are made up of only one large exon, not sharing common 3' exons as expected. These neural cadherin-like cell adhesion proteins are integral plasma membrane proteins. Their specific functions are unknown but they most likely play a critical role in the establishment and function of specific cell-cell neural connections.

Glypican-1 (GPC1), glypican-2 (GPC2) and glypican-3 (GPC3) are members of the glypican family of heparan sulfate proteoglycans, which attach to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor. Cell-surface heparan sulfate proteoglycans participate in molecular events that regulate cell adhesion, migration, and proliferation. Glypican-2, a cell surface proteoglycan bearing heparan sulfate, may have a function related to the motile behaviors of developing neurons. Ligation of cell-surface glypican-2 with midkine (MK) or an; against epitope-tagged glypican-2 induces cell adhesion and promotes neurite outgrowth. MK binds to heparan sulfate chains of glypican-2 in a manner similar to syndecan-3, but different localization of epitope-tagged glypican-2 and syndecan-3 on the surface of N2?cells suggests that they may play different roles in MK-mediated neural function.
