Description:
Cyclins, regulatory subunits, which associate with kinases, control many of the important steps in cell cycle progression. The Cdc2 protein kinase (p34Cdc2) exhibits protein kinase activity in vitro and exists in a complex with both cyclin B and a protein homologous to p13SUC1. Cdc2 kinase is the active subunit of the M phase promoting factor (MPF) and the M phase-specific Histone H1 kinase. The p34Cdc2/cyclin B complex is required for the G2 to M transition. An additional cell cycle-dependent protein kinase, termed p55cdc, exhibits a high degree of homology with the S. cerevisiae proteins Cdc20 and Cdc4. The p55cdc transcript is readily detectable in a variety of cultured cell lines in growth phase, but disappears when cell growth is chemically arrested.

PLAG1 is a transcription factor whose activation results in upregulation of target genes, such as IGFII, leading to uncontrolled cell proliferation: when overexpressed in cultured cells, higher proliferation rate and transformation are observed. Other target genes such as CRLF1, CRABP2, CRIP2, PIGF are strongly induced in cells with PLAG1 induction. Proto oncogene whose ectopic expression can trigger the development of pleomorphic adenomas of the salivary gland and lipoblastomas. Overexpression is associated with upregulation of IGFII, is frequently observed in hepatoblastoma, common primary liver tumor in childhood. Cooperates with CBFB-MYH11, a fusion gene important for myeloid leukemia.

The Pax gene family of nuclear transcription factors is comprised of nine members that function during embryogenesis to regulate the temporal and position-dependent differentiation of cells. In addition, the family is involved in a variety of signal transduction pathways in the adult organism. Mutations in the Pax family of proteins have been linked to disease and cancer in humans. Pax-7 is a protein specifically expressed in cultured satellite cell-derived myoblasts. In situ hybridization reveals that Pax-7 is also expressed in satellite cells residing in adult muscle. A chromosomal aberration in the gene encoding Pax-7 causes rhabdomyosarcoma 2 (RMS2) (also called alveolar rhabdomyosarcoma).

The rTS gene codes for a naturally occurring antisense RNA to thymidylate synthase (TS) mRNA and two proteins (rTSalpha and rTSbeta (also known as ENOSF1)). The role of the major protein product of rTS, ENOSF1 has been linked to alterations in TS protein expression, but the precise function of ENOSF1 is unknown. Expression of rTS is associated with growth arrest in cell culture, but its overexpression has been noted in cells resistant to anticancer drugs such as 5-fluorouracil and methotrexate. Studies have shown that increased expression of ENOSF1 is associated with the decrease in TS protein expression due to production of novel, diffusible signal molecules.

Fibroblast growth factor-1 also designated basic FGF, are members of a family of growth factors that stimulate proliferation of cells of mesenchymal, epithe-lial and neuroectodermal origin. Additional members of the FGF family include the oncogenes FGF-3 (Int2) and FGF-4 (hst/Kaposi), FGF-5, FGF-6, FGF-7 (KGF), FGF-8 (AIGF), FGF-9 (GAF) and FGF-10–FGF-23. Members of the FGF family share 30-55% amino acid sequence identity and similar gene structure, and are capable of transforming cultured cells when overexpressed in transfected cells. Cellular receptors for FGFs are members of a second multigene family including four tyrosine kinases, designated Flg (FGFR-1), Bek (FGFR-L), TKF and FGFR-3.
