9 is a transmembrane glycoprotein that is selectively expressed on the cell surface of B-lymphocytes,where it activates intracellular signaling cascades involving both Ras and phosphatidylinositol 3-kinase pathways.Lymphocytes proliferate and differentiate in response to various concentrations of different antigens. The ability of the B cell to respond in a specific, yet sensitive manner to the various antigens is achieved with the use of low-affinity antigen receptors. This gene encodes a cell surface molecule which assembles with the antigen receptor of B lymphocytes in order to decrease the threshold for antigen receptor-dependent stimulation.

The cellular homolog of v-Myb, designated c-Myb, encodes a transcription factor. The CD137 ligand (CD137L, 4-1BBL) is a member of the tumor necrosis factor (TNF) family. CD137L is a type II, transmembrane protein found on activated macrophages, dendritic cells and mature B cells. The interaction with its receptor 4-1BB induces recruitment of TNF receptor-associated factor 1 (TRAF1) and TRAF2 and interaction with the kinase p56lck. CD137 and CD137L have been reported to be involved in tumor rejection, apoptosis, anti-viral immunity, diabetes, in T and B cell co-stimulation and modulation of the immune response.

Non-receptor protein-tyrosine kinase that regulates reorganization of the actin cytoskeleton, cell polarization, cell migration, adhesion, spreading and bone remodeling. Plays a role in the regulation of the humoral immune response, and is required for normal levels of marginal B-cells in the spleen and normal migration of splenic B-cells. Required for normal macrophage polarization and migration towards sites of inflammation. Regulates cytoskeleton rearrangement and cell spreading in T-cells, and contributes to the regulation of T-cell responses. Promotes osteoclastic bone resorption; this requires both PTK2B/PYK2 and SRC. May inhibit differentiation and activity of osteoprogenitor cells.
