The 53-7.3 monoclonal antibody is specific for mouse CD5, a 67 kDa protein that is expressed on most thymocytes, mature T cells and the B-1 subset of B cells. CD5 is a member of the scavenger receptor cysteine-rich protein superfamily and is a ligand for the c-type lectin CD72. CD5 is also known as Leu-1 and functions to modulate antigen receptor signaling on T and B cells.

Expression of LASS2 is transiently increased during the period of active myelination and is specifically localized to white matter tracts of the brain, including the Schwann cells of sciatic nerves suggesting that LASS2 is important for the synthesis of dihydroceramide used for synthesis of myelin sphingolipids. Expression of LASS2 in hepatocellular carcinoma cell lines suppresses the growth of cancer cells.

CD17 is an intermediate glycosphingolipid from the metabolism of higher gangliosides that localizes to sphingolipid-sterol rafts. CD17 is detectable in monocytes, granulocytes, basophils, platelets, a subset of peripheral B cells (CD19 ) and tonsil dendritic cells. It is rapidly down regulated on activated granulocytes and is upregulated on IL-2 activated T lymphocytes. CD17 binds to bacteria and may function in phagocytosis. VEGF-treated endothelial cells can produce CD17, which can then mediate signaling toward PECAM-1 expression and angiogenesis. Tumor necrosis factor alpha (TNFa)-induced astrogliosis (astrocyte proliferation and glial fibrillary acidic protein (GFAP) upregulation) in response to neuro-inflammation (i.e. spinal cord injury) causes an increase in intracellular levels of CD17. Aberrant levels of glycosphingolipids are a feature of cancer cells and may influence integrin clustering and internalization.

The coiled-coil domain is a structural motif found in proteins that are involved in a diverse array of biological functions such as the regulation of gene expression, cell division, membrane fusion and drug extrusion and delivery. CCDC42 (coiled-coil domain containing 42) is a 316 amino acid protein encoded by a gene that maps to human chromosome 17p13.1. Encoding over 1,200 genes, chromosome 17 comprises over 2.5% of the human genome. Two key tumor suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Tumor suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome.

As a subfamily of the cadherin superfamily, protocadherins are cadherin-like cell adhesion proteins that contain up to seven extracellular domains and are predominantly expressed in the nervous system. Importantly, the adhesion mechanism of protocadherins is distinct from classic cadherins. Through inactivation or overexpression, several protocadherins have been implicated in a variety of cancers. Protocadherin-20 (PCDH20), also known as protocadherin-13, is a 924 amino acid protein containing 6 cadherin domains and potentially functioning as a calcium-dependent cell-adhesion protein. In non-small cell lung cancer cell lines, a homozygous loss of PCDH20 was identified through either deletion of one allele and methylation of the other or methylation of both alleles. Hypermethylation of PCDH20 is associated with worse prognosis and clinical outcome, suggesting that PCDH20 may function as a tumor suppressor.
