The PDE4A8 is a testis germ cell-specific PDE4A variant. There are two PDE4A variants identified in rat testis (PDE4A8 and an un-characterized 76 kDa PDE4A protein). Spermatogenesis is accompanied by expression of a varied repertoire of PDE4 enzymes at different stages of cell differentiation that presumably perform specialized functions. Confocal microscopic examination revealed the presence of PDE4A8 in primary and secondary spermatids but not in mature spermatozoa. PDE4A8 inhibition results in increased apoptosis of the germ cells accompanied with reduction in the number of mature spermatozoa.

TNFRSF14 is a type I membrane protein belonging to the TNF receptor superfamily. This receptor mediates herpes virus entry into cells during infection. TNFRSF14 is able to inhibit the proliferation, activation, and cytokine production of T cells. It has an extracellular domain containing several cysteine-rich repeats and a short cytoplasmic region containing a TRAF (TNF receptor-associated factor) interaction domain. The extracellular domain of TNFRSF14 interacts with the herpes simplex virus envelope glycoprotein D. TNFRSF14 binds two cellular ligands: lymphotoxin alpha and LIGHT. LIGHT is a transmembrane protein expressed and shed from the surface of activated T cells, exhibits inducible expression, and competes with HSV glycoprotein D for HVEM, a receptor expressed by T lymphocytes. The LIGHT:TNFRSF14 interaction controls immune response functions by cell death induction as well as cell activation. TNFRSF14 is expressed by peripheral blood T cells, B cells, monocytes and in various tissues enriched in lymphoid cells.

The 2F1 antibody reacts with mouse KLRG1 (Killer cell Lectin-like Receptor G1). This 30-38 kDa homodimeric receptor may be expressed by activated, mature NK cells and by effector/memory T cells, with potentially different roles in each cell type. KLRG1 can regulate, in an inhibitory fashion, the development and effector functions of NK cells, and is often cited as a senescence or terminal differentiation marker for T cells. Ligands for KLRG1 include members of the cadherin family of adhesion molecules, specifically N-Cadherin, E-Cadherin, and R-Cadherin. These interactions may induce bidirectional, immunosuppressive signaling in both KLRG- and Cadherin-expressing cells. A more recently identified role for KLRG1-Cadherin signaling in tissue organization, e.g. in cardiac angiogenesis, expands the function of these interactions beyond immunosuppression of immune cells. CD135 is also expressed on malignant hematopoietic cells including AML, ALL and CML BC. CD135, also known as FMS-like tyrosine kinase 3, FLT3, STK1, and Flk2, is a growth factor receptor that binds the FLT3 ligand to promote the growth and differentiation of primitive hematopoietic cells. The intracytoplasmic domain of CD135 is modified by phosphorylation and has been shown to interact with Grb2, SOCS1, VAV1, and Shc. In humans, expression of Flt3 is restricted to subsets of CD34 positive as well as CD34 negative normal bone marrow cells. In these cells, the level of expression of Flt3 is rather low.
