Description:
A variety of morphological and molecular changes are required for mature spermatozoa formation. These steps are temporally guided by the transcription and translation of several testis-specific genes. SPANX (sperm protein associated with the nucleus, X-linked) family members are sperm- and testis-specific proteins containing between 97-103 amino acids, whose genes form a cluster on chromosome X. Sharing a high level of sequence similarity, SPANX-A, -B, -C, -D and -E localize to both cytoplasm and nucleus where they are associated with nuclear craters. SPANX-B (Sperm protein associated with the nucleus on the X chromosome B/F) is a 103 amino acid protein that is detected in round and elongating spermatids.

Description:
TSKS is a 592 amino acid protein that is highly expressed in human testicular tissue. Low levels of TSKS are detectable in prostate, placenta, fetal liver, thymus, and mammary gland tissues. TSKS is found to be downregulated in cancerous testicular tissue from seminoma, teratocarcinoma, embryonal and Leydig cell tumours concurrently with high expression in neighboring premalignant carcinoma. TSKS protein contains an N-terminal signal peptide, but does not contain a transmembrane region. TSKS has many potential phosphorylation and glycosylation sites and is phosphorylated by soluble recombinant TSSK2 in vitro. It is thought that TSKS likely plays a physiological role in spermatogenesis or spermiogenesis.

Comparative study of this MAb and a standard CD68 MAb showed that their antigens are different. This MAb shows a restricted reactivity to cells of the monocyte/macrophage system. It specifically reacts with blood monocytes and stains resident macrophages in a wide variety of human tissues. This MAb does not stain antigen-presenting cells, e.g., Langerhans cells. Reportedly, its reactivity is restricted to histiocytes and macrophages.

Recognizes a protein of 12 kDa, identified as β-microglobulin. β-microglobulin non-covalently associates with the 44 kDa chain to form the HLA Class I antigen complex. Human β-2 microglobulin associated with HLA Class I antigens is expressed on many types of cells including lymphocytes, thymocytes, monocytes, granulocytes, platelets, endothelial cells, and epithelial cells. It is absent on erythrocytes. This MAb is specific to human β-2 microglobulin and does not react with non-human primate cells. This antibody reacts with all cell types excluding erythrocytes. The detection of β-2 microglobulin in body fluids has been used as a tumor marker and for monitoring patients with HIV infection.

Propidium Iodide (PI) is a membrane-impermeant DNA binding dye that cannot penetrate viable cells. PI rapidly enters cells with compromised membranes and intercalates between base pairs allowing exclusion of non-viable cells from analysis of flow cytometry data. BAMBI is a membrane spanning glycoprotein that acts as a negative regulator of TGF-beta signaling during development. The BAMBI family of proteins are related to type I TGF-beta receptor family, however, BAMBI is a pseudoreceptor that lacks an intracellular serine/threonine kinase domain. BAMBI transcription regulation is under the influence of beta-catenin, BMP, smad3 and smad4. BAMBI expression can increase in colorectal and hepatocellular carcinomas relative to non-cancerous tissues. BAMBI is expressed at high levels during odontogenesis. BAMBI is coexpressed with Bmp-4 during early Xenopus embryogenesis and can be detected in poorly metastatic human melanoma cell lines.

Recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. This MAb is a useful marker for melanocytes and melanomas.

Uroplakin-3A (UPK3A) is a component of the asymmetric unit membrane (AUM). It is a highly specialized bio-membrane made by terminally differentiated urothelial cells. The protein may play an important role in AUM-cytoskeleton interaction in terminally differentiated urothelial cells. UPK3A also contributes to the formation of urothelial glycocalyx, which may play an important role in preventing bacterial adherence through FimH bacterial protein binding leading to bladder infection. UPK3A has been shown as a helpful marker for the detection of bladder cancer.
