Description:
The Oxysterol-binding protein (OSBP) family of proteins consist of OSBP (OSBP1) and OSBP2 (ORP-4), which share a high overall similarity. OSBPs are involved in lipid metabolism and signal transduction, as well as vesicle transport, and can translocate to the periphery of Golgi membranes when they are bound to oxysterols. The OSBP protein transports sterols from lysosomes to the nucleus, where sterols downregulate the genes for HMG synthetase, HMG-CoA reductase and the low density lipoprotein receptor (LDLR). OSBP localizes to the cytosol and is widely expressed, while OSBP2 is mainly detected in testis, retina and fetal liver. The extracellular signal-regulated kinase (ERK) signaling pathway is controlled by OSBP via its cholesterol-binding properties. OSBP binds with a high affinity to 25-hydroxy-cholesterol (25-HC), a suppressor of cholesterol synthesis gene transcription in cultured cells.

Recognizes a 53 kDa protein, which is identified as p53 suppressor gene product. It reacts with the mutant as well as the wild form of p53 under denaturing and non-denaturing conditions. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Anti-p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intratubular germ cell neoplasia. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.

Humanin, an endogenous anti-apoptotic peptide against Alzheimer disease-related insults, consists of 24 amino acids. The secreted protein is a neuroprotective factor against death induced by several different types of Alzheimer's disease genes. Humanin protects neuronal cells from damage caused by Alzheimer's disease genes, specifically APP (amyloid precursor protein). Humanin acts as a ligand for formyl peptide receptor-like 1 (FPRL1) with APP and utilizes its neuroprotective effects by inhibiting FPRL1 access to APP. The peptide prevents Bax translocation from cytosol to mitochondria. Humanin expression levels may be dependent on defects in energy production in muscles with mitochondrial abnormalities. The peptide has been detected in muscles of patients with the mitochondrial disease chronic progressive external ophthalmoplegia (CPEO). Humanin is mainly expressed in the kidney, heart, liver, testis and skeletal muscles.

The Decorin core protein binds to growth factors, intercellular matrix molecules such as Fibronectin and Throm-bospondin, and to the Decorin endocytosis receptor. Fibromodulin is a collagen-binding keratan sulphate proteoglycan that influences adhesion processes of connective tissue and plays a role in fibrillogenesis by regulating collagen fibril spacing and thickness. Keratocan (KTN) develops corneal transparency and maintains the stromal matrix structure. Keratocan is a secreted protein in the extracellular matrix that binds to keratan sulfate chains. Keratocan is mainly detected in the cornea, but can also be expressed in trachea, intestine, ovary, lung and skeletal muscle. Defects in the gene encoding for Keratocan can cause cornea plana 2 (CNA2), an autosomal recessive disorder where the forward convex curvature of the cornea is flattened.

This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

Description:
Capases are a highly conserved family of cysteine proteases and are synthesized as inactive zymogens, which are cleaved at Asp-x sites during apoptosis, generating large and small subunits that heterodimerize to form the active enzyme. Caspase-1 is a member of the ICE/CED-3 family of caspases and is detected in the pro-form as a protein of 45 kDa.
Caspase-1 generates the bioactive form of the pro-inflammatory cytokine IL-1β from its biologically inactive precursor. Overexpression of caspase-1 in cells induces apoptosis, which can be inhibited by cytokine response modifier A (CrmA). Caspase-1 mRNA is predominantly expressed in the spleen and lung and to a lesser extent in skeletal muscle, kidney, testis and heart and within these tissues macrophages preferentially express caspase-1. Caspase-1 deficient mice have no gross abnormalities, however, macrophages from caspase-1-deficient mice are defective in IL-β processing and release.

IGSF1 is a highly glycolylated immunoglobulin domain-containing protein. IGSF1 has been shown to act as a coreceptor in inhibin signaling, however, it does not appear to be a high-affinity inhibin receptor by itself. May reduce or inhibit activin A signaling and is believed to be necessary in the mediation of specific effects of inhibin B on activin-stimulated transcription. IGSF1 has been found to interact with several members of the ACVR family and possibly some members of the BMPR group. There are 3 known isoforms of IGSF1, with 1 and 2 likely being multi-pass membrane proteins. Isoform 3 is believed to be expressed as a secreted form. Expression is high in pancreas, testis and fetal liver, while heart, prostate and small intestine show only moderate expression. IGSF1 may be found at very low levels in brain, muscle, thymus, ovary, colon, fetal lung and fetal kidney. Isoform 3 has been detected in pituitary gland.
