Non-receptor protein-tyrosine kinase that regulates reorganization of the actin cytoskeleton, cell polarization, cell migration, adhesion, spreading and bone remodeling. Plays a role in the regulation of the humoral immune response, and is required for normal levels of marginal B-cells in the spleen and normal migration of splenic B-cells. Required for normal macrophage polarization and migration towards sites of inflammation. Regulates cytoskeleton rearrangement and cell spreading in T-cells, and contributes to the regulation of T-cell responses. DENND4C is a 1673 amino acid multi-pass membrane protein. DENND4C contains a dDENN domain, DENN domain, uDENN domain and a PPR (pentatricopeptide) repeat. As a result of alternative splicing, six DENND4C isoforms exist. The gene encoding DENND4C maps to human chromosome 9, which houses over 900 genes and comprises nearly 4% of the human genome. Hereditary hemorrhagic telangiectasia, which is characterised by harmful vascular defects, and Familial dysautonomia, are both associated with chromosome 9. Notably, chromosome 9 encompasses the largest interferon family gene cluster.

Gelsolin is a calcium dependent actin binding protein. It is a potent modulator of actin filament length and gelation. Gelsolin has been shown to exist in at least two variant forms, cytoplasmic gelsolin and plasma gelsolin. Plasma gelsolin has also been called actin depolymerizing factor (ADF) or brevin. Human plasma and rabbit macrophage gelsolins differ by the presence of a 25 amino acid residue extension at the NH2 terminus in the human plasma gelsolin, which appears to account for the difference in relative molecular weights.

Apoptosis is mediated by death domain containing adapter molecules and a caspase family of proteases. Certain serine/threonine protein kinases, such as ASK1 and RIP, are mediators of apoptosis. Two novel serine/threonine kinases that induce apoptosis were recently identified and designated DRAK1 and DRAK2 (for DAP kinase related apoptosis inducing protein kinases). DRAKs contain an N terminal kinase domain and a C terminal regulation domain. Overexpression of DRAK2 induces apoptosis. DRAKs have high sequence homology to DAP and ZIP kinases, and they represent a novel family of serine/threonine kinases, which mediates apoptosis through their catalytic activities. In Xenopus retinal photoreceptor cells, the rhythmic regulation of Nocturnin is thought to be controlled by phosphorylated CREB. Mice lacking Nocturnin remain lean on high fat diets with a reduction in visceral fat, which suggests that this protein may also be responsible for lipid metabolism and fat storage.
