Description:   Zinc-finger proteins bind nucleic acids and play important roles in various cellular functions, including cell proliferation, differentiation, and apoptosis. This protein and Grb10-interacting GYF protein 2 have been identified as a components of the mammalian 4EHP (m4EHP) complex. The complex is thought to function as a translation repressor in embryonic development. [provided by RefSeq, Oct 2012].

Description:   Proteins containing PDZ domains have been shown frequently to bind the C-termini of transmembrane receptors or ion channels. They have also been shown to bind to other PDZ domain proteins and could possibly be involved in intracellular signalling. PDZK3 contains six PDZ domains and shares sequence similarity with pro-interleukin-16 (pro-IL-16). Like pro-IL-16, the encoded protein localizes to the endoplasmic reticulum and is thought to be cleaved by a caspase to produce a secreted peptide containing two PDZ domains. In addition, the PDZK3 gene is upregulated in primary prostate tumors and may be involved in the early stages of prostate tumorigenesis.

Description:   HD-PTP is a 1636 amino acid protein encoded by the human gene PTPN23. HD-PTP belongs to the protein-tyrosine phosphatase family, non-receptor class subfamily. It contains one BRO1 domain, two TPR repeats and one tyrosine-protein phosphatase domain. The C-terminal region contains the PTP-like domain, whereas the N-terminal region contains the two TPR regions. These regions are homologous to the yeast protein, BRO1, which is involved in the mitogen-activated protein kinase signaling pathway. Similarly, HD-PTP is believed to act as a negative regulator of Ras-mediated mitogenic activity and is phosphorylated upon DNA damage, probably by ATM or ATR. HD-PTP protein is differentially modulated by two angiogenic growth factors. While Vascular Endothelial Growth Factor (VEGF) has no affect on protein levels, Fibroblast Growth Factor-2 (FGF-2) induces HD-PTP degradation via the proteasome system.

Description:   The ProSAP family of proteins contain many protein-protein interaction domains and serve as scaffolding mediators within the post-synaptic density (PSD) of excitatory brain synapses. The PSD is an electron-dense structure underneath the post-synaptic plasma membrane of excitatory synapses that anchors and clusters glutamate receptors opposite to the pre-synaptic neurotransmitter release site. Shank proteins contain PDZ modular domains that coordinate the synaptic localization of ion channels, receptors, signaling enzymes, and cell adhesion molecules. The PDZ domain mediates protein-protein interactions via the recognition of a conserved sequence motif at the C-terminus of their target protein(s). ProSAPiP1 (proline rich synapse associated protein interacting protein 1) is a 673 amino acid protein that interacts with the PDZ domain of Shank 3. ProSAPiP1 expression is brain-specific with highest expression within the cerebellum, hippocampus and cerebral cortex.

Description:   Zinc-finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. ZDHHC20 (zinc finger, DHHC-type containing 20) is a 365 amino acid multi-pass membrane protein that contains one DHHC-type zinc finger and is thought to function as a palmitoyltransferase, catalyzing the transformation of palmitoyl-CoA and a cysteine-conjugated protein to an S-palmitoyl protein and free CoA. In response to DNA damage, ZDHHC20, which exists as multiple alternatively spliced isoforms, may be phosphorylated by ATM or ATR. The gene encoding ZDHHC20 maps to human chromosome 13, which houses over 400 genes, such as BRCA2 and RB1, and comprises nearly 4% of the human genome.

Description:   SAP 155 is a 1,304 amino acid member of the SF3B1 family and contains eleven HEAT repeats. Localized to nuclear speckles and also to the cytoplasm during mitosis, SAP 155 is a subunit of the SF3B splicing factor. The SF3B splicing factor is a U2 snRNP-associated protein complex essential for spliceosome assembly. SF3B contains the spliceosomal proteins SAP 49, SAP 130, SAP 145 and SAP 155. Concomitant with splicing catalysis, SAP 155 is phosphorylated at its N-terminal Thr-Pro dipeptide motifs by Dyrk1A and cyclin E/Cdk2. This modification of SAP 155 is vital for a functional spliceosome as it is an essential event in the basic splicing reaction. Due to alternative splicing events, various SAP 155 isoforms are produced.

Description:   Acts as a cofactor for complement factor I, a serine protease which protects autologous cells against complement-mediated injury by cleaving C3b and C4b deposited on host tissue. May be involved in the fusion of the spermatozoa with the oocyte during fertilisation. Also acts as a costimulatory factor for T-cells which induces the differentiation of CD4+ into T-regulatory 1 cells. T-regulatory 1 cells suppress immune responses by secreting interleukin-10, and therefore are thought to prevent autoimmunity. A number of viral and bacterial pathogens seem to exploit this property and directly induce an immunosuppressive phenotype in T-cells by binding to CD46.

Description:   Thrombopoietin (TPO or THPO), also known as c-Mpl ligand (c-Mpl L), is a cytokine that plays a central role in megakaryopoiesis by influencing the development and maturation of megakaryocytes and platelet production from hematopoietic stem cells. TPO exerts its biological effects through the TPO receptor, c-Mpl. c-Mpl is a member of the cytokine receptor superfamily. Expression of c-Mpl is restricted to hematopoietic tissues and cells, such as bone marrow, spleen, fetal liver and CD³⁴⁺ cells. Stimulation of c-Mpl with TPO results in the activation of the Janus tyrosine kinase family members, Tyk 2 and JAK2, which in turn phosphorylate Stat5 and Stat3, causing their nuclear translocation and the transcription of Stat responsive genes. Mutations in c-Mpl have been implicated as the cause of certain human disorders, including congenital amegakaryocytic thrombocytopenia (CAMT) and thrombocytopenia with absent radii (TAR) syndrome.

Description:   Produces nitric oxide (NO) which is implicated in vascular smooth muscle relaxation through a cGMP-mediated signal transduction pathway. NO mediates vascular endothelial growth factor (VEGF)-induced angiogenesis in coronary vessels and promotes blood clotting through the activation of platelets. May play a significant role in normal and abnormal limb development.

Description:   This gene encodes the acidic form of complement factor 4, part of the classical activation pathway. The protein is expressed as a single chain precursor which is proteolytically cleaved into a trimer of alpha, beta, and gamma chains prior to secretion. The trimer provides a surface for interaction between the antigen-antibody complex and other complement components. The alpha chain may be cleaved to release C4 anaphylatoxin, a mediator of local inflammation. Deficiency of this protein is associated with systemic lupus erythematosus and type I diabetes mellitus. This gene localizes to the major histocompatibility complex (MHC) class III region on chromosome 6. Varying haplotypes of this gene cluster exist, such that individuals may have 1, 2, or 3 copies of this gene. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2011].
